维生素D_(3)对酒精性肝损伤小鼠的保护作用

摘要 目的探讨维生素D_(3)对酒精性肝损伤小鼠的保护作用。方法40只小鼠随机分为4组:正常对照组(Control组)、单纯维生素D_(3)组(VitD_(3)组)、酒精组(EtOH组)及酒精+维生素D_(3)组(EtOH+VitD_(3)组)。采用Lieber-DeCarli酒精液体饲料喂养小鼠制备酒精性肝损伤模型,检测血清中丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)水平以及肝指数,HE染色观察肝脏病理形态学改变,qRT-PCR检测肿瘤坏死因子α(TNF-α)、转化生长因子β(TGF-β)、白细胞介素(IL)-6和IL-1β的mRNA相对表达水平,Western blot检测肝脏NF-κB p65和NF-κB p50蛋白表达。结果与Control组比较,EtOH组小鼠血清ALT、AST活力、肝指数及肝组织TNF-α、TGF-β、IL-6和IL-1βmRNA水平显著升高,EtOH组肝细胞索排列紊乱,肝小叶界限不清,肝细胞出现明显炎性细胞浸润和脂肪空泡,NF-κB p65和NF-κB p50蛋白表达水平显著升高;与EtOH组比较,EtOH+VitD_(3)组小鼠血清ALT、AST活力、肝指数及肝组织TNF-α、TGF-β、IL-6和IL-1βmRNA水平显著降低,且病理染色结果显示肝细胞炎性细胞浸润和脂肪空泡数量减少,肝脏恢复正常细胞结构,同时NF-κB p65和NF-κB p50蛋白表达水平显著降低。结论维生素D_(3)对酒精性肝损伤小鼠具有一定的保护作用,其机制可能与抑制NF-κB信号通路,降低机体炎症反应有关。 Objective To investigate the protective effect of vitamin D_(3)on alcoholic liver injury in mice.Methods Forty mice were randomly divided into 4 groups:normal Control(Control)group,vitamin D_(3)(VitD_(3))group,alcohol model(EtOH)group and alcohol+vitamin D_(3)(EtOH+VitD_(3))group.The mice were fed with the DeCarli alcohol liquid diet to establish alcoholic liver injury model.Serum levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST)and liver index were detected.HE staining was used to observe the pathological changes of liver.The relative expression levels of tumor necrosis factorα(TNF-α),transforming growth factorβ(TGF-β),interleukin-6(IL-6)and interleukin-1β(IL-1β)mRNA were detected by quantitative real-time PCR(qRT-PCR).The expressions of nuclear factor-kappa B(NF-κB)p65 and NF-κB p50 in liver were detected by Western blot.Results The serum ALT,AST vitality,liver index and hepatic TNF-α,TGF-β,IL-6 and IL-1βmRNA in EtOH group were significantly higher than those in Control group.EtOH group disorganized hepatocyte and hepatic lobules boundary was not clear,and the hepatocytes showed apparent inflammatory cells infiltration of liver cells and fat cavitation.NF-κB p65 and NF-κB p50 protein expression increased significantly.Compared with EtOH group,the serum ALT,AST vitality,liver index and hepatic TNF-α,TGF-β,IL-6 and IL-1βmRNA in EtOH+VitD_(3)group decreased significantly.The pathological staining results showed that inflammatory cells infiltration and decrease in the number of fat vacuoles,and the liver cells returned to normal liver cell structure.At the same time the NF-κB p65 and NF-κB p50 protein expression level decreased significantly.Conclusion Vitamin D_(3)has a certain protective effect on alcohol-induced liver injury in mice,and its main mechanisms are related to the inhibition of NF-κB pathway and the reduction of inflammatory response.
