摘要
为研究两种糖水平下α-硫辛酸(α-LA)对日本沼虾肠道菌群的影响,以玉米淀粉为糖源,配制2×3因子的试验饲料,即2种糖水平(15%和30%)、3种α-LA浓度(0、700和1400 mg/kg),将平均体重为(0.050±0.003) g的1 500尾日本沼虾幼虾随机分为6组,每组5个平行,进行为期8周的饲养试验.结果表明:在相同α-LA下,30%糖水平组日本沼虾肠道菌群的Chao指数和Ace指数显著低于15%糖水平组(P<0.05),但在30%糖水平下,随着α-LA的增加,Chao指数和Ace指数显著升高(P<0.05);日本沼虾肠道菌群主要以厚壁菌门(Firmicutes)、变形菌门(Proteobacteria)、软壁菌门(Tenericutes)和放线菌门(Actinobacteria)4个门为主,糖水平和α-LA浓度对其丰度具有显著的交互作用(P<0.05),在15%糖水平下,摄食700 mg/kg α-LA组的变形菌门和软壁菌门的相对丰度显著高于其余两组(P<0.05);从属水平上分析,日本沼虾肠道菌群主要以Tyzzerella_3属、肠杆菌属(Enterobacter)、红球菌属(Rhodococcus)、气单胞菌属(Aeromonas)和假单胞菌属(Pseudomonas)5个属为主,肠杆菌属主要受α-LA浓度的影响,而气单胞菌属主要受糖水平影响.由此可见,高糖能降低肠道菌群的丰度,α-LA的添加可缓减高糖对肠道菌群相对丰度的抑制作用,在两种糖水平下添加700 mg/kg α-硫辛酸,可改善日本沼虾肠道菌群的结构和稳定性.
In order to investigate the effects of alpha lipoic acid(α-LA)on the intestinal microflora of oriental river prawn Macrobrachium nipponense,six diets were formulated using corn starch as the carbohydrate source,at two carbohydrate levels(15%and 30%)and three concentrations ofα-LA supplementation(0,700,and 1400 mg/kg).Each experimental diet was fed to prawns(mean weight 0.050±0.003 g),twice daily to apparent satiation,in five replicates.The feeding trial lasted for 8 weeks.The results are as follows:(1)At the sameα-LA concentration,the Chao and Ace indexes in 30%(high)-carbohydrate groups were significantly lower than that of 15%(low)-carbohydrate groups.But the Chao and Ace indexes increased with the increase ofα-LA in the 30%(high)-carbohydrate groups.(2)At the phylum level,there were 4 dominant bacteria phyla in the intestinal tract,including Firmicutes,Proteobacteria,Tenericutes and Actinobacteria.There was significant interaction between the carbohydrate andα-LA levels and abundance of gut microflora(P<0.05).Under 15%carbohydrate level,the abundance of Proteobacteria and Tenericutes in prawns fed 700 mg/kgα-LA was significantly higher than that of other two groups,while under 30%carbohydrate level,the abundance of Firmicutes in prawns fed 700 mg/kgα-LA was significantly higher than that of other two groups.(3)At the genus level,there were 5 dominant bacteria genera in the intestinal tract,including Tyzzerella_3,Enterobacter,Rhodococcus,Aeromonas and Pseudomonas.Enterobacter was mainly affected byα-LA concentration,while Aeromonas was mainly affected by carbohydrate level.Thus,α-LA supplementation can alleviate the negative effect of reduced abundance by high carbohydrate,and 700 mg/kgα-LA supplementation under either dietary carbohydrate level could improve the stability of intestinal florain of M.nipponense.
