摘要
近年来基因编辑技术发展迅速,规律成簇的间隔短回文重复序列及相关蛋白9(Clustered regularly interspaces short palindromic repeats and CRISPR-associated protein 9,CRISPR/Cas9)作为一种效率高、成本低和操作简单的新型基因编辑技术脱颖而出,不论是在动植物还是在微生物方面都被广泛应用。作为重要的家禽,鸡的生殖结构特点限制其在遗传育种中的研究。CRISPR/Cas9技术以操作精准、便利的优势扭转了传统基因编辑技术在鸡遗传育种研究中难以应用的局面。文章就CRISPR/Cas9技术的发展和作用机制及其在鸡生长发育、性腺分化、疾病防控、胚胎编辑、转基因鸡制备等方面的最新研究进展进行了综述,同时也对CRISPR/Cas9技术的脱靶及改良应用进行了简要论述,为CRISPR/Cas9技术在鸡标准化、规模化养殖过程中的实际运用提供参考。
Gene editing technology has developed rapidly in recent years and as a novel gene editing technology with high efficiency,low cost and easy operation,clustered regularly interspaced short palindromic repeats and CRISPR-associated protein 9(CRISPR/Cas9)stands out among a host of traditional technologies in both plants and animals as well as in microorganisms.As important poultry,chicken is limited in genetic breeding research due to their unique reproductive structural characteristics.CRISPR/Cas9 has reversed the difficulty of applying traditional gene editing techniques in chicken genetic breeding research with the advantages of precise and convenient operation.This study reviewed the development and mechanism of action of CRIS⁃PR/Cas9 technology and its latest research progress in chicken growth and development,gonadal differentiation,disease preven⁃tion and control,embryo editing and preparation of transgenic chickens,and also briefly discussed the off-target and improved ap⁃plications of CRISPR/Cas9 technology,which would play an important role in the practical application of CRISPR/Cas9 technolo⁃gy in chicken standardization and large-scale breeding.The study also briefly discussed on the off-target and improved application of CRISPR/Cas9 technology,which could play an important role in the practical application of CRISPR/Cas9 technology in standardized and large-scale chicken breeding.
