骨质疏松与冠状动脉钙化相关机制研究进展 被引量:2

摘要 骨质疏松与冠状动脉粥样硬化性心脏病是两类常见的老年疾病,都以高发病率、残疾率和死亡率为特征。既往认为骨质疏松与冠状动脉钙化相互独立,近年来多项研究证明两者在病理机制方面存在联系。NOSIP、GXYLT2、TRIM63、COL1A1、THBS1、SR-BI、VKORC1等基因及非编码RNA不仅与骨质疏松有关,而且参与血管钙化的发生发展。核因子κB受体激活蛋白/核因子κB受体激活蛋白配体/护骨因子、Klotho基因/成纤维细胞生长因子23、β联蛋白等分子信号通路以及骨调节蛋白如骨形态发生蛋白、胎球蛋白及钙磷代谢紊乱在介导骨质疏松与血管钙化的病理过程中具有重要作用。通过研究两者的共同发病机制对共同预防和治疗具有重要意义,本文就两者在遗传基因、分子信号通路、临床相关研究等进行综述,为骨质疏松和心血管疾病的早期预防提供依据。 Osteoporosis and cardiovascular disease are two common health concerns in the elderly,both characterized by a high incidence rate,disability and mortality.It was previously considered that osteoporosis and coronary artery calcification were independent of each other.However,a number of recent studies have shown that there is a relationship between the two in the pathological mechanism.Genes including NOSIP,GXYLT2,TRIM63,COL1A1,THBS1,SR-BI,VKORC1 and non-coding RNA are not only related to osteoporosis,but also involved in the occurrence and development of vascular calcification.Molecular signaling pathways such as receptor activator of nuclear factor-κB/receptor activator of nuclear factor-κB ligand/osteoprotegerin,Klotho protein/fibroblast growth factor-23,Wnt/β-catenin and bone regulatory proteins such as bone morphogenetic protein,Fetuin-A,calcium and phosphorus metabolism disorders play an important role in mediating the pathological process of osteoporosis and vascular calcification.It is of great significance for the joint prevention and treatment of two diseases by studying the common pathogenesis.Therefore,this study reviewed the two from genetic genes,molecular signaling pathways and clinical studies,so as to provide a basis for the early prevention of osteoporosis and cardiovascular diseases.
