摘要
【目的】探明不同药剂浸种及营养液培养浓度对魔芋实生种子水培生长的影响,以期筛选出最优的浸种药剂及最佳的营养液培养浓度应用于生产。【方法】以“秦魔1号”魔芋实生种子为材料,进行不同药剂浓度溶液(硫脲0.50%,1.00%;KNO30.20%,0.50%;GA 1 mg/L,5 mg/L;α-萘乙酸50 mg/L,100 mg/L)浸种处理;参考日本园试通用营养液配方配制标准营养液,将催芽后的种子置于不同浓度的营养液(100%标准营养液,80%标准营养液,60%标准营养液)中培养。测定分析不同药剂浸种处理魔芋的萌发率及魔芋幼苗的生长情况(株高、叶幅、叶幅/株高、生根数、根长、球茎重量、球茎膨大倍数),筛选魔芋幼苗培养的最佳营养液浓度。【结果】不同药剂浸种处理除100 mg/L α-萘乙酸外,均对魔芋实生种子的萌发起促进作用,以1.0%硫脲和5 mg/L GA处理促萌发效果较优,发芽率分别为78%和68%。不同营养液浓度对魔芋实生种子的生长都有一定促进作用,以标准营养液对魔芋实生种子处理作用效果最佳,株高(14.9 cm)、叶幅(21.2 cm)、叶幅/株高(1.42)、生根数(16条)、根长(19.7 cm)、球茎重(13.14 g)、球茎膨大倍数(38.54倍)均最高。【结论】魔芋实生种子浸种药剂选择1.0%硫脲和5 mg/L GA较好,生长营养液以100%标准营养液最佳。
【Objective】The effect of different chemical soaking and nutrient solution concentration on water culture and growth of konjac seed is studies,so as to screen the optimal chemical and concentration of nutrient solution for production.【Method】The konjac seed“Qinmo 1”are treated with different chemical concentration(0.50%thiourea,1.00%thiourea,0.20%KNO 3,0.50%KNO 3,1 mg/L GA,5 mg/L GA,50 mg/Lα-naphthylacetic acid,100 mg/Lα-naphthylacetic acid);According to the Japanese garden test general formula to make up the standard nutrient solution,which is used to culture the bud-induced seed with different concentration(100%standard solution,80%standard solution,60%standard solution).The germination rate and growth status(plant height,leaf width,leaf width/plant height,the number of root,root length,protocorm weight,the expansion of protocorm)under different chemical soaking are determined and analyzed,and the best nutrient solution concentration is screened.【Results】Except 100 mg/Lα-naphthylacetic acid,other chemical-soaking treatment all have prompting function to the germination of konjac seedling,and 1.0%thiourea and 5 mg/L GA have better germination effect,whose germination rate are 78%and 68%,respectively.Different nutrient solution concentration all have prompting effect on the growth of konjac seed,among which the standard nutrient solution has the best effect on the seed,as the plant(14.9 cm),leaf width(21.2 cm),leaf width/plant height(1.42),the number of root(16),root length(19.7 cm),protocorm weight(13.14 g),the expansion of protocorm(38.54 times)are the highest.【Conclusion】1.0%thiourea and 5 mg/L GA should be chosen as seed-soaking chemicals for konjac seed,and the 100%standard nutrient solution are the optimal growth nutrient solution.
