摘要
天然中药在抗肿瘤方面具有独特优势,而细胞焦亡途径的发现为中医药靶点的选择提供了新方向。随着对细胞焦亡认识的深入及其与肿瘤治疗相关性的探索,诱导细胞焦亡以促进化疗、免疫治疗等疗法更好地发挥抑制肿瘤增殖及瘤体生长作用机制的研究愈加深入。中医阴阳平衡理论对细胞焦亡的有序、适度控制具有重要指导意义,而中医药因其多靶点、多途径特点对肿瘤化疗及免疫治疗增敏、减少耐药性的优势逐渐显现。但目前中药有效成分靶向细胞焦亡途径的研究尚处于基础阶段,且以单味药单成分为主,尚未明确靶向机制及上下游靶点;中药复方相关研究较少,且其发挥细胞焦亡作用的主要成分并不明确。针对此,今后可结合网络药理学、分子对接等生物信息技术或基因敲除方式对中药各成分影响细胞焦亡的机制进行多维度、多靶位及多因子分析,并在中医平衡理论及“损其有余,补其不足”治疗原则的指导下引导适度可控的焦亡。
Natural Chinese medicine has unique advantages in anti-tumor,and the discovery of the pyroptosis pathway provides a new direction for the selection of Chinese medicine targets.With the in-depth understanding of pyroptosis and the exploration of its correlation with tumor therapy,the research on the mechanism of inducing pyroptosis to promote chemotherapy,immunotherapy,and other therapies to better inhibit tumor proliferation and tumor growth has become more and more mature.The theory of TCM Yin-Yang balance has important guiding significance for the orderly and moderate control of pyroptosis,and the TCM advantages in enhancing the sensitivity of tumor chemotherapy and immunotherapy and reducing drug resistance due to its multi-target and multi-channel characteristics are gradually emerging.However,at present,the research on pyroptosis pathways targeted by TCM active ingredients is still in the basic stage,and it is mainly a single ingredient of a single drug,and the targeting mechanism and upstream and downstream targets have not yet been clarified.And there are few relevant studies on Chinese herbal compound prescriptions,and the main components that play a role in pyroptosis are not clear.Given this,in the future,network pharmacology,molecular docking,and other bioinformatics technologies or gene knockout methods can be combined to conduct multi-dimensional,multi-target,and multi-factor analyses on the mechanism of each TCM component affecting pyroptosis.And guide moderate and controllable pyroptosis under the guidance of TCM balance theory and treatment principles"Cutting what is more than enough,making up what is not enough".
