摘要
对大鼠灌胃浙贝母后的尿液、粪便样品预处理方法进行筛选,并对其经预处理和未经预处理冻存条件下的稳定性进行考察.采用UPLC-Q-TOF-MS技术建立了不同的预处理方法的含药和空白尿液与粪便的图谱,对5种尿液样品预处理方法和4种粪便样品预处理方法进行考察筛选,并对尿液和粪便样品经预处理和未经预处理冻存0、3、5、7、10、15 d后的稳定性进行考察.研究结果表明,乙腈沉淀蛋白法为尿液和粪便样品的最佳预处理方法,经前处理和未经前处理冻存15 d的含药尿液和粪便样品稳定性良好.本实验为后续尿液与粪便中的成分分析及体内代谢研究奠定了基础.
The pretreatment methods of urine and feces samples from Fritillaria thunbergii after gavage in rats were screened,and their stability after pretreatment and non-pretreatment frozen storage conditions was investigated.The chromatograms of drug-containing and blank urine and feces with different pretreatment methods were established using UPLC-Q-TOF-MS technology.Five urine sample pretreatment methods and four feces sample pretreatment methods were investigated and screened,and the stability of urine and feces samples after pretreatment and non-pretreatment frozen for 0,3,5,7,10 and 15 days was investigated.The results showed that acetonitrile precipitation protein method was the optimal pretreatment method for urine and feces samples,and the stability of drug-containing urine and feces samples stored for 15 days after pretreatment and without pretreatment was good.This study laid the foundation for subsequent component analysis in urine and feces and in vivo metabolism research.
