摘要
目的采用生物信息学方法,分析儿童克罗恩病的热点基因、功能及免疫状态。方法通过GEO(gene expression omnibus)数据库的GSE126124数据集,筛选克罗恩病儿童及正常儿童的信息,选择组织样本作为研究对象。共下载了37例克罗恩病儿童患者的病历作为实验组,19例正常儿童作为正常组。比较克罗恩病肠道组织和正常肠道组织的差异基因;对差异基因进行功能富集分析、通路富集分析及蛋白-蛋白网络分析;通过R语言分别计算2组受试者的免疫评分,采用t检验进行比较。结果差异基因分析获得120个基因,包含了87个上调基因及33个下调基因;京都基因和基因组数据库(kyoto eacyclopedia of genes and genomes,KEGG)通路主要富集于细胞因子-细胞因子受体相互作用(cytokine-cytokine receptor interaction)、白介素-17信号通路(IL-17 signaling pathway)和NOD样受体信号通路(NOD-like receptor signaling pathway)。蛋白-蛋白网络分析筛选出27个热点基因。免疫评分提示实验组患者13个主要免疫基因的表达情况高于正常组。结论通过生物信息学初步揭开了儿童克罗恩病的潜在热点基因、信号通路及免疫状态,为后续进一步研究提供了基础。
Objective To analyze the hotspot genes,function and immune status of Crohn’s disease in children using a bioinformatics approach.Methods The GSE126124 dataset of GEO(gene expression omnibus)database was used to screen the information of children with Crohn’s disease and normal children,and tissue samples were selected as the study subjects.A total of 37 medical records of pediatric Crohn’s disease patients were downloaded as the experimental group and 19 normal children as the normal group.Differential genes were compared by comparing Crohn’s disease intestinal tissues with normal intestinal tissues.Functional enrichment analysis,pathway enrichment analysis and protein-protein association analysis were performed on the differential genes.The immune scores of the 2 groups of subjects were calculated separately by R language and compared by t-test.Results Differential gene analysis obtained 120 genes,containing 87 up-regulated genes and 33 down-regulated genes.The kyoto encyclopedia of genes and genomes(KEGG)pathway was mainly enriched in cytokine-cytokine receptor interactions,interleukin-17 signaling pathway and NOD-like receptor signaling pathway.Protein-protein network analysis screened out 27 hotspot genes.The immune score suggested that the expression profile of 13 major immune genes was higher in the experimental group than in the normal group.Conclusion Bioinformatics has initially uncovered potential hotspot genes,signaling pathways and immune status in childhood Crohn’s disease,which provides the basis for further studies to follow.
