摘要
探讨鸡源肠外致病性大肠埃希氏菌(ExPEC)生物被膜(BF)形成能力及BF与耐药性之间的相关性,采用96孔微量板法和正交试验测定10株鸡源ExPEC的BF形成能力和BF强阳性菌株的最佳成膜条件,使用二倍倍比稀释法测定β-内酰胺类抗生素对生物被膜态与浮游态鸡源ExPEC的最小抑菌浓度(MIC)。结果显示,10株鸡源ExPEC的BF阳性率为90%,E-26、E-28、E-43为弱阳性菌株(+);E-27、E-30、E-42为中等阳性菌株(++),E-39、E-40、E-41为强阳性菌株(+++),E-20为阴性菌株;3株强阳性菌株BF最佳成膜条件不相同;3株BF强阳性鸡源ExPEC的BF形成后,对β-内酰胺类抗生素耐药性有不同程度增加,同种抗生素对不同菌株的MIC变化可能不同,不同抗生素对同一菌株变化也不同。因此,鸡源ExPEC的BF阳性率较高,但各菌株形成BF的能力存在差异,不同菌株形成BF所需的条件不完全相同,BF形成会使鸡源ExPEC对抗生素的耐药水平增强,但同为BF阳性鸡源ExPEC,形成BF后β-内酰胺类抗生素耐药水平变化存在差异。
The experiment was to investigate the ability of extraintestinal pathogenic Escherichia coli(ExPEC)to form biofilm(BF)and the correlation between BF and drug resistance.The BF-forming ability of 10 chicken-derived ExPEC strains and the optimal film-forming conditions of BF-positive strains were determined by 96-well microplate method and orthogonal test.The minimum inhibitory concentration(MIC)of planktonic chicken-derived ExPEC toβ-lactam antibiotics was determined by two-fold dilution method.The results showed that the BF positive rate of 10 chicken-derived ExPEC was 90%,E-26,E-28,E-43 were weakly positive strains(+);E-27,E-30,E-42 were moderately positive strains(++),E-39,E-40,E-41 were strong positive strains(+++),E-20 was negative strain;the best film-forming conditions of BF of the three strongly positive strains are different;After the formation of BF of strongly positive chicken-derived ExPEC,the resistance toβ-lactam antibiotics increased to different degrees.The MIC changes of the same antibiotic against different strains may be different,and the changes of different antibiotics against the same strain were also different.Therefore,the BF positive rate of chicken-derived ExPEC is high,but the ability of each strain to form BF is different,and the conditions required for different strains to form BF are not exactly the same.The formation of BF will increase the resistance level of chicken-derived ExPEC to antibiotics,but The same BF-positive chicken-derived ExPEC showed differences in the changes ofβ-lactam antibiotic resistance after the formation of BF.
