基于酶底物法对饮用水和非饮用水中嗜肺军团菌的定量测定

摘要 研究采用酶底物法对饮用水和非饮用水中的嗜肺嗜肺军团菌进行定量检测。结果表明,酶底物法对检测嗜肺嗜肺军团菌的质控样品有较高的准确度和稳定性。对饮用水实际样品中嗜肺军团菌进行检测,在30份管网水中未检出阳性结果,在60份二次供水样品中检出率达5%,其中3份阳性样品均来自公共场所建筑自来水龙头,阳性样品的检出浓度为1.0~5.2MPN/(100mL)。对非饮用水实际样品中嗜肺军团菌进行检测,在全部30份水样中检出率达67%,阳性样品的检出浓度为2.2~1992.6MPN/(100mL)。对酶底物法检测出的阳性样品均进行了菌落验证试验,无假阳性现象,表明酶底物法检测饮用水和非饮用水中实际水样的嗜肺军团菌特异度为100%。其结果表明,在非饮用水和饮用水末端公共场所区域水体中,仍存在嗜肺军团菌污染风险,对人群构成潜在威胁。 In this study,the enzyme substrate method was used for quantitative detection of Legionella pneumophila in drinking and non-drinking water.The enzyme substrate method has high accuracy and stability for the detection of Legionella pneumophila in quality control samples.No Legionella pneumophila was detected in 30 water samples from drinking water supply networks and 5%of 60 secondary water supply samples,three of the positive samples were from tap water in public buildings.The detection concentration of the positive samples ranged from 1.0 MPN/(100 mL)to 5.2 MPN/(100 mL).The detection rate of Legionella pneumophila in non-potable water samples was 67%,and the detection range of positive samples was 2.2 MPN/(100 mL)to 1992.6 MPN/(100 mL).All the positive samples detected by enzyme substrate method were tested by colony assay,and no false positive was found.The results showed that the Legionella pneumophila specificity of enzyme substrate method for detecting actual water samples in drinking and non-drinking water was 100%.The results suggested that there is still a risk of Legionella pneumophila contamination in non-drinking water and in the water bodies of public places at the end of drinking water,posing a potential threat to the population.
