摘要
目的:探讨Ⅰ型胶原α2链(COL1A2)在胃癌中的表达与免疫细胞浸润的相关性及其对患者预后的影响。方法:通过TCGA、TIMER数据库分析了不同肿瘤组织中COL1A2的基因表达水平及胃癌组织与正常组织中COL1A2的基因表达差异;利用TIMER数据库分析了COL1A2对胃癌患者预后及肿瘤免疫细胞浸润的影响;进一步采用Kaplan-Meier plotter数据库分析COL1A2与胃癌患者预后的关系,通过单因素Cox回归和构建列线图模型分析COL1A2对胃癌预后的评估价值;GSEA富集分析COL1A2可能参与的分子通路;通过TCGA数据库分析COL1A2基因表达与免疫细胞浸润的相关性。结果:COL1A2在胃癌组织中显著高表达(P<0.001),COL1A2与T分期、临床分期和组织学分级具有相关性(P<0.05);COL1A2高表达的胃癌患者生存率低于低表达患者,且COL1A2可作为胃癌患者的独立预后因子(P<0.05)。GSEA富集分析显示COL1A2主要富集在轴突引导通路、细胞黏附分子通路等。TCGA数据库分析显示COL1A2与免疫细胞浸润具有显著相关性。结论:COL1A2在胃癌患者中特异性高表达,且与免疫细胞浸润具有显著相关性,COL1A2有望成为胃癌免疫细胞浸润和预后相关的生物学标志物。
Objective:To investigate the correlation between the expression of typeⅠcollagenα2 chain(COL1A2)in gastric cancer and the infiltration of immune cells and its influence on the prognosis of patients.Methods:The gene expression levels of COL1A2 in different tumor tissues and the gene expression difference of COL1A2 in gastric cancer tissues and normal tissues were analyzed by TCGA and TIMER databases.The effects of COL1A2 on the prognosis of gastric cancer patients and tumor immune cell infiltration were analyzed by TIMER.Kaplan-Meier plotter was utilized to figure out the relation between COL1A2 and prognosis of gastric cancer patients.Univariate Cox regression and construction of nomogram model were used to analyze the prognostic value of COL1A2 in gastric cancer.GSEA enrichment was used to analyze the molecular pathways that COL1A2 may be involved in.The correlation between COL1A2 gene expression and immune cell infiltration was analyzed by TCGA database.Results:COL1A2 was significantly expressed in gastric cancer tissues(P<0.001),and COL1A2 was correlated with T stage,clinical stage and histological grade(P<0.05).The survival rate of GC patients with high COL1A2 was lower than that with low COL1A2,and COL1A2 can be used as an independent prognostic factor in gastric cancer patients(P<0.05).GSEA enrichment analysis showed that COL1A2 was mainly enriched in axon guidance pathway and cell adhesion molecule pathway.TCGA database analysis showed that COL1A2 was significantly associated with immune cell infiltration.Conclusion:COL1A2 is specifically highly expressed in gastric cancer patients,and has a significant correlation with immune cell infiltration.COL1A2 is expected to be a biomarker related to immune cell infiltration and prognosis in gastric cancer.
