摘要
目的探讨结直肠癌组织中尾型同源盒基因2(cadual homeobox gene 2,CDX2)、缺氧诱导因子(hypoxia inducible factor,HIF)-1α蛋白表达情况及肿瘤芽孢间的相关性。方法回顾性收集西安交通大学第一附属医院2012年1月至2015年9月期间符合本研究纳入条件的63例手术切除的结直肠癌标本,利用免疫组织化学链霉菌抗生物素蛋白-过氧化物酶连接两步法检测CDX2和HIF-1α蛋白表达,光学显微镜下观察染色情况及肿瘤芽孢并对肿瘤芽孢分级,利用Spearman等级相关性检验分析三者之间的相关性。结果CDX2和HIF-1α在结直肠癌组织中的蛋白阳性表达者分别为35例(55.6%)和47例(74.6%),二者的蛋白阳性表达呈负相关(r_(s)=–0.302,P=0.017);CDX2和HIF-1α在结直肠癌组织中芽孢部位的蛋白阳性表达者分别为21例(51.2%)和26例(63.4%),二者的蛋白阳性表达亦呈负相关(r_(s)=–0.336,P=0.031),但未发现肿瘤芽孢分级与结直肠癌组织中CDX2或HIF-1α蛋白阳性表达之间有相关性(r_(s)=0.113,P=0.370;r_(s)=–0.026,P=0.838)。结论从本研究结果看,同一结直肠癌标本及其内芽孢部位中CDX2和HIF-1α蛋白阳性表达均呈负相关,未发现肿瘤芽孢分级与结直肠癌组织中CDX2、HIF-1α蛋白阳性表达之间有关,提示在结直肠癌恶性进展过程中缺氧环境可能参与了CDX2水平的下调。
Objective To investigate the correlations among the cadual homeobox gene 2(CDX2),hypoxia inducible factor-la(HIF-1a)protein expressions,and tumor budding in the colorectal cancer(CRC).Methods In this study,63 CRC specimens surgically removed in the First Affiliated Hospital of Xi'an Jiaotong University from January 2012 to September 2015 were collected.The CDX2 and HIF-1a protein expressions were detected by immunohistochemical staining streptavidin-biotin peroxidase two-step method.The staining and the grade of tumor budding were observed under an optical microscope,and the correlation was analyzed using Spearman test.Results The positive expressions of CDX2 and HIF-la proteins in the CRC tssues were 35(55.6%)and 47(74.6%)cases,respectively,which was a negative correlation in the CRC(r_(s)=-0.302,P=0.017).The positive expressions of CDX2 and HIF-1αproteins in the tumor budding of colorectal cancer were 21(51.2%)and 26(63.4%)cases,respectively,which was also a negative correlation in the tumor budding of CRC(r_(s)=-0.336,P=0.031),but there was no statistic correlation between the grade of tumor budding and CDX2 or HIF-1αpositive protein expression in the CRC(r_(s)=0.113,P=0.370;r_(s)=-0.026,P=0.838).Conclusions The positive expression between CDX2 and HIF-1a has a negative correlation in the same CRC specimen and which has a negative correlation in tumor budding.There is no statistic correlation between grade of tumor budding and CDX2 or HIF-la protein expression in the CRC.Hypoxia environment may be involved in the downregulation of CDX2 level during the malignant progression of CRC.
