内异膏对子宫内膜异位症大鼠子宫内膜容受性的影响

Effect of Neiyi Ointment on Endometrial Receptivity in Rats with Endometriosis

摘要 【目的】探讨内异膏(活血补肾为主中药组成)对子宫内膜异位症大鼠的治疗作用及机制。【方法】采用自体移植法构建子宫内膜异位症大鼠模型。实验分成假手术组7只,模型组6只,内异膏低、中、高剂量组各8只,阿司匹林组5只,连续给药28 d。在动情期取材,测量子宫内膜异位病灶体积、质量,采用苏木素-伊红(HE)染色法观察子宫内膜组织病理形态,扫描电镜观察在位内膜胞饮突,荧光定量聚合酶链反应(PCR)检测大鼠子宫在位内膜同源框基因10(HOXA10)mRNA表达,免疫组织化学法检测子宫内膜整合素αvβ3、白血病抑制因子(LIF)表达。【结果】(1)与模型组比较,内异膏中、高剂量组及阿司匹林组大鼠子宫内膜异位病灶质量、体积明显减小(P<0.05),内异膏低剂量组异位病灶质量、体积无明显改变(P>0.05)。(2)与模型组比较,内异膏各剂量组与阿司匹林组子宫内膜异位内膜组织均有不同程度萎缩,其中,内异膏中、高剂量组及阿司匹林组效果更明显。(3)假手术组子宫内膜胞饮突表达多处于成熟期,模型组胞饮突处于发育期,内异膏各剂量组胞饮突较模型组发育有所改善,成熟的胞饮突有所增多,阿司匹林组成熟胞饮突增多更明显。(4)与假手术组比较,模型组大鼠子宫在位内膜组织HOXA10 mRNA表达水平差异无统计学意义(P>0.05);与模型组比较,内异膏中剂量组大鼠子宫在位内膜组织HOXA10 mRNA表达水平降低(P<0.05),而其余治疗组差异无统计学意义(P>0.05)。(5)各组子宫内膜组织整合素αvβ3和LIF表达比较,差异均无统计学意义(P>0.05)。【结论】内异膏可改善子宫内膜异位症大鼠胞饮突发育,使成熟期胞饮突数量增多,提高子宫内膜容受性。 Objective To investigate the therapeutic effect and mechanism of Neiyi Ointment in the treatment of rats with endometriosis.Methods An endometriosis rat model was constructed by autologous transplantation.There were a sham-operation group of 7,a model group of 6,and Neiyi Ointment low-,medium-and high-dose group(8 in each group),and an Aspirin group of 5 in the experiment.During the motility phase the samples were taken,the volume and mass of endometriotic lesions were measured.The endometrial histopathology was observed by hematoxylin-eosin(HE)staining,the cytosolic protrusion of the in situ endometrial tissue was observed by scanning electron microscopy,the mRNA expression of the homologous endometrial frame gene 10(HOXA10)was detected by fluorescence quantitative polymerase chain reaction(PCR),and the expressions of endometrial integrinαvβ3 and leukemia inhibitory factor(LIF)were detected by immunohistochemistry.Results(1)Compared with the model group,the volume and mass of endometriotic lesions in the rats in the medium-and high-dose groups of Neiyi Ointment and the Aspirin group were significantly reduced(P<0.05),while the volume and mass of endometriotic lesions in the low-dose group of Neiyi Ointment did not significantly changed,the differences being not statistically significant(P>0.05).(2)Compared with the model group,the endometriotic tissues in each dose group of the Neiyi Ointment and the Aspirin group had different degrees of atrophy.(3)The expression of endometrial pinopodes in the sham-operation group was mostly in the mature stage,while the endometrial pinopodes in the model group were in the developmental stage.The development of endometrial pinopodes in each dose group of endoisol was improved compared with the model group,the mature endometrial pinopodes being increased,and the increase of mature endometrial pinopodes in the Aspirin group was more obvious.(4)Compared with the sham-operation group,the difference in the mRNA expression level of HOXA10 in the in situ endometrial tissue of rats in the model group was not statistically significant(P>0.05);compared with the model group,the mRNA expression level of HOXA10 in the in situ endometrial tissue of rats in the medium-dose group of Neiyi Ointment was reduced(P<0.05),while the difference in the remaining treatment groups was not statistically significant(P>0.05).(5)The differences in the expression of integrinαvβ3 and LIF in the endometrial tissue of each group were not statistically significant(P>0.05).Conclusion The Neiyi Ointment improved the development of endometrial pinopodes in rats with endometriosis,increasing the number of endometrial pinopodes at maturity and thus improving endometrial tolerance.
