摘要
基于液相色谱-质谱(LC-MS)法和网络药理学方法选取茗酿养生酒的功能活性成分及其作用靶点,获取其和高脂血症疾病的交集靶点,构建“活性成分-交集靶点-疾病”网络图,使用STRING数据库进行蛋白互作(PPI)网络分析,并使用Metascape数据库进行基因本体论(GO)和京都基因与基因百科全书(KEGG)通路富集分析,研究茗酿养生酒中功能活性成分影响高脂血症的作用机制。结果表明,从茗酿养生酒中筛选得到21种活性成分,其与高脂血症有92个交集靶点,关键靶点包括肿瘤坏死因子(TNF)、白细胞介素-6(IL-6)和血管内皮生长因子(VEGFA)等,其涉及对细菌的响应过程等1 100个GO条目和糖尿病并发症中的晚期糖基化终产物-糖基化终末产物受体(AGE-RAGE)信号通路等120条KEGG信号通路,说明茗酿养生酒中的功能成分影响高脂血症具有多成分-多靶点-多途径的作用特点。
The functional active components and their action targets of Mingniang health liquor were selected based on LC-MS and network pharmacology method,the intersection targets with hyperlipidemic diseases were obtained,and the network diagram of"active ingredients-intersection targets-diseases"was constructed.Protein-protein interaction(PPI)network analysis was performed using STRING database,the gene ontology(GO)and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis were carried out by Metascape database,and the effect mechanism of the functional active ingredients in Mingniang health liquor on hyperlipidemia was studied.The results showed that 21 active components were screened from the Mingniang health liquor,which had 92 intersection targets with hyperlipidemia,key targets included tumor necrosis factor(TNF),interleukin-6(IL-6),vascular endothelial growth factor(VEGFA)and so on.It involved 1100 GO entries such as the response process to bacteria and 120 KEGG signaling pathways such as the advanced glycosylated end-product receptor(AGE-RAGE)signaling pathway in diabetic complications.These results indicated that the functional components in Mingniang health liquor affect hyperlipidemia with the characteristics of multi-component,multi-target and multi-pathway.
