基于指纹图谱与多指标含量测定的黔产香榧种子质量控制研究

摘要 目的建立黔产香榧种子高效液相色谱(HPLC)的指纹图谱及多指标含量测定方法,为黔产香榧种子的质量控制与开发提供依据。方法采用Agilent Eclipse Plus C 18色谱柱(4.6 mm×150 mm,5μm);以乙腈(A)-0.06%醋酸-水溶液(B)为流动相,梯度洗脱;流速1.0 mL/min;检测波长280 nm;柱温30℃;进样量15μL。结果20批香榧种子指纹图谱相似度均大于0.983,共确定5个共有峰,指认了没食子酸和绿原酸。对样品中的没食子酸和绿原酸2种化学成分进行含量测定与方法学考察,包含专属性、线性、精密度、重复性和稳定性,均符合要求;平均回收率在97.75%~98.82%,RSD≤1.62%。香榧种子中该2种化学成分的含量分别为33.25~97.00μg/g和1072.28~1279.56μg/g。结论本研究所建立黔产香榧种子的HPLC指纹图谱和多指标含量测定的方法准确可行,可作为黔产香榧种子质量评价的一种有效手段。 Objective To establish fingerprinting and multi-component content determination methods by high performance liquid chromatography(HPLC)of the Torreya grandis seed,and provide the scientific basis for quality control and development of Torreya grandis seed.Methods Agilent Eclipse Plus C 18 column(4.6 mm×150 mm,5μm)was used.With acetonitrile(A)-0.06%acetic acid-water solution(B)as mobile phase,gradient elution was performed.The flow rate was 1.0 ml/min.The detection wavelength was 280 nm.The column temperature was 30℃.The injection volume was 15μL.Results The similarity of 20 batches of Torreya grandis seed fingerprints were all greater than 0.983.A total of 5 common peaks were identified,and gallic acid and chlorogenic acid were identified.The methodological investigations were carried out on the determination of gallic acid and chlorogenic acid in the samples,including specificity,linearity,precision,repeatability,and stability,and all met the requirements.The average recoveries ranged from 97.75%to 98.82%,RSD≤1.62%.The contents of these two chemical components in Torreya grandis seed were 33.25-97.00μg/g and 1072.28-1279.56μg/g,respectively.Conclusion The method established by this research is accurate and feasible,and can be used as an effective means for quality control of Torreya grandis seed.
