摘要
目的:探讨在牙周膜成纤维细胞(hPDLFs)中上调LncRNA-KAT7对脂多糖诱(LPS)诱导的白介素-17(IL-17)表达的影响。方法:纳入慢性牙周炎患者24例,健康志愿者17例为对照组。采集实验组和对照组的临床数据并采用qRT-PCR检测两组牙龈组织中LncRNA-KAT7和IL-17的表达水平,分析两者之间的相关性;分离培养hPDLFs,检测不同来源的hPDLFs中LncRNA-KAT7和IL-17的表达;使用LPS刺激hPDLFs,并过表达LncRNA-KAT7,观察炎症因子IL-17表达的变化。结果:在慢性牙周炎组织中,LncRNA-KAT7呈低表达,IL-17呈高表达,且两者为负相关性;不同组织来源的hPDLFs未经LPS处理时,LncRNA-KAT7和IL-17的表达无明显差异;未经LPS处理,过表达LncRNA-KAT7对IL-17的表达无统计学差异,但LPS处理后,上调LncRNA-KAT7可明显抑制IL-17的表达。结论:在hPDLFs中上调LncRNA-KAT7可抑制LPS诱导炎症因子IL-17的表达。
Objective:To investigate the effects of overexpression of LncRNA-KAT7 on the expression of LPS-induced IL-17 expression in human periodontal ligament fibroblasts(hPDLFs).Methods:24 patients with chronic periodontitis were included in the test group,and 17 healthy volunteers in the control group.The clinical data of the subjects were collected and the expression levels of LncRNA-KAT7 and IL-17 in the gingival tissues of the 2 groups were detected by QRT-PCR,and the correlation between LncRNA-KAT7 and IL-17 was analyzed.Then,hPDLFs were isolated and cultured,the expression of LncRNA-KAT7 and IL-17 in hPDLFs were detected.Finally,hPDLFs were treated with LPS following LncRNA-KAT7 upregulation,the expression of IL-17 was observed.Results:Low expression of LncRNA-KAT7 and high expression of IL-17 were observed in chronic periodontitis tissues,and the two were negatively correlated.The expression of LncRNA-KAT7 and IL-17 in hPDLFs of the test group without LPS treatment showed no significant difference compared with that of the control group.Without LPS treatment,overexpression of LncRNA-KAT7 had no effect on IL-17 expression,but after LPS treatment,up-regulation of LncRNA-KAT7 significantly inhibited the expression of IL-17.Conclusion:Upregulation of LncRNA-KAT7 in hPDLFs can inhibit the expression of LPS-induced IL-17.
