摘要
目的检测微小RNA-137(miR-137)和FAM98A在子宫内膜癌(EC)组织中的表达,并探究其与患者临床病理特征及预后的关系。方法以郑州市妇幼保健院收治的85例EC患者为研究对象,收集各研究对象EC组织、匹配的癌旁正常组织及临床病理特征资料。应用实时荧光定量PCR(qRT-PCR)检测组织中miR-137、FAM98A mRNA表达水平,免疫组化染色检测组织中FAM98A蛋白表达;Pearson相关分析EC组织中miR-137与FAM98A mRNA表达水平相关性;2检验分析EC组织中miR-137、FAM98A表达与患者临床病理特征的关系;Kaplan-Meier法分析EC组织中miR-137、FAM98A表达与患者预后生存的关系。结果与癌旁正常组织相比,EC组织中miR-137表达水平显著降低(1.02±0.13 vs.0.58±0.10,P<0.001),而FAM98A mRNA表达水平及FAM98A蛋白阳性表达率显著升高(1.06±0.17 vs.2.85±0.26、23.53%vs.61.18%,P<0.001);EC组织中miR-137与FAM98A mRNA表达水平呈负相关(r=-0.663,P<0.001);EC组织中miR-137和FAM98A表达与患者肿瘤分化程度、国际妇产科联盟(FIGO)分期、肌层浸润深度、淋巴结转移有关(P<0.05);miR-137高表达组EC患者3年累积生存率显著高于miR-137低表达组(83.33%vs.60.47%,P=0.018),而FAM98A阳性表达组EC患者3年累积生存率显著低于FAM98A阴性表达组(63.46%vs.84.85%,P=0.033)。结论EC组织中miR-137呈低表达,而FAM98A呈高表达,且二者均与患者肿瘤分化程度、FIGO分期等临床病理特征及预后密切相关,可能成为EC新的生物标志物。
Objective To assess the expression of microRNA-137(miR-137)and FAM98A in endometrial carcinoma(EC)tissues,and to investigate their correlation with clinicopathological characteristics and prognosis of patients.Methods From January 2017 to January 2019,85 EC patients admitted to Zhengzhou Maternal and Child Health Hospital were enrolled.The EC tissues,and matched adjacent normal tissues were collected.The expression levels of miR-137 and FAM98A mRNA in tissues were assessed by real-time quantitative PCR(qRT-PCR).The protein expression of FAM98A in tissue was assessed by immunohistochemical staining.The correlation between miR-137 and FAM98A mRNA expression levels in EC tissues was analyzed by Pearson correlation.The correlations between the expression of miR-137 and FAM98A in EC tissue and the clinicopathological characteristics of patients were analyzed by2 test.And the correlation between the expression of miR-137 and FAM98A in EC tissues and the prognosis and survival of patients was analyzed by Kaplan-Meier method.Results Compared with adjacent normal tissues,the expression level of miR-137 in EC tissues significantly decreased(1.02±0.13 vs.0.58±0.10,P<0.001);however,the expression level of FAM98A mRNA and the positive expression rate of FAM98A protein were significantly increased(1.06±0.17 vs.2.85±0.26,23.53%vs.61.18%,P<0.001).The expression levels of miR-137 and FAM98A mRNA in EC tissues were negatively correlated(r=-0.663,P<0.001).The expression of miR-137 and FAM98A in EC tissues was significantly correlated to the degree of tumor differentiation,the International Federation of Gynecology and Obstetrics(FIGO)stage,the depth of myometrial invasion and lymph node metastasis(P<0.05).The 3-year cumulative survival rate of EC patients in the high miR-137 expression group was significantly higher than that in the low miR-137 expression group(83.33%vs.60.47%,P=0.018),while the 3-year cumulative survival rate of EC patients in the FAM98A positive expression group was significantly lower than that in the FAM98A negative expression group(63.46%vs.84.85%,P=0.033).Conclusion The expression of miR-137 in EC tissues is low,while the expression of FAM98A is high,and both are closely correlated to the clinicopathological characteristics(tumor differentiation,FIGO stage,and etc.)and prognosis of patients,and may become new biomarkers for EC.
