摘要
在河北省唐山市滦南县林场,以高0.5~0.8 m、地径(1.5±0.2)cm的3年生银杏(Ginkgo biloba)实生苗(邳州的梅核品种)为砧木,以直径(1.0±0.1)cm的优株枝条为接穗(6个品系:13#、16#、27#、叶丰、大花穗、泰兴大佛指)进行嫁接,每个无性系各嫁接60~90株;试验采用随机区组设计,共设3个区组,作为试验的3个重复,每个区组包括6个小区,6个银杏品系随机分配在各小区;在每个小区的植株上方、距地面高2 m,搭建2种不同透光度的遮阴网(遮光率为25%、45%的遮阴),以不遮阴作为对照,试验设3个重复,每个重复包括20株苗木;以6个品系的3年生银杏嫁接苗为试验材料,测定净光合速率、酶活性、次生代谢物、总黄酮醇苷,分析不同遮阴处理对6个银杏品系叶净光合速率、酶活性、总黄酮质量分数的影响,遴选有利于银杏黄酮积累的最适光照强度、最优银杏品系。结果表明:银杏净光合速率,随着光照强度的降低而呈现降低的趋势;遮光率为25%的遮阴处理时苯丙氨酸酶(PAL)活性最高,遮光率为25%的遮阴处理、遮光率为45%的遮阴处理有利于银杏肉桂酸-4-羟基化酶(C4H)活性增加,遮光率为25%的遮阴处理时泰兴大佛指苯丙氨酸酶、肉桂酸-4-羟基化酶活性最大(分别为15.79、43.80 U·g^(-1));银杏次生代谢物槲皮素、山奈酚、异鼠李素、总黄酮醇苷质量分数,在遮光率为25%的遮阴处理时较高,其中泰兴大佛指总黄酮醇苷质量分数最高(达到11.28 mg·g^(-1));相关性分析表明,苯丙氨酸酶、肉桂酸-4-羟基化酶活性,与总黄酮醇苷质量分数都呈现显著正相关。综合试验结果,泰兴大佛指最适合作为叶用品种在唐山地区栽培,并且遮光率为25%的遮阴处理更能促进其黄酮的积累。
Three-year-old Ginkgo biloba grafted seedlings of 6 strains were used as experimental materials.The light intensities were controlled as L1(no shading),L2(25%shading)and L3(45%shading),respectively,by covering shading nets.The changes of net photosynthetic rate,enzyme activity and total flavonol glycoside content of G.biloba leaves were analyzed to explore the optimal light intensity and the best strain for the accumulation of ginkgo flavonoids.The results showed that the net photosynthetic rate of G.biloba decreased with the decrease of light intensity;The activity of PAL enzyme was the highest under L2 treatment,and L2 and L3 treatments was conducive to the increase of C4H enzyme activity of G.biloba,and the PAL and C4H enzyme activities of Taixing Buddha Finger were the highest under L2 treatment,which were 15.79 and 43.80 U·g^(-1) respectively.The contents of quercetin,kaempferol,isorhamnetin and total flavonol glycosides in G.biloba were higher under L2 treatment,among which the content of total flavonol glycosides in Taixing Buddha Finger was the highest,reaching 11.28 mg·g^(-1).Correlation analysis showed that PAL and C4H enzyme activities were significantly positively correlated with the content of total flavonol glycosides.Comprehensive analysis shows that Taixing Buddha Finger is the most suitable leaf variety for cultivation in Tangshan,and 25%shading treatment can promote the accumulation of flavonoids.
