miR-183调控Wnt/β-catenin信号通路促进非小细胞肺癌特性肿瘤干细胞及上皮间质转化 被引量:2

miR-183 regulates Wnt/β-catenin signaling pathway to promote tumor stem cell and epithelial mesenchymal transformation in non-small cell lung cancer

摘要 目的研究miR-183在非小细胞肺癌中的表达情况及影响非小细胞肺癌的作用机制。方法收集10例新鲜非小细胞肺癌及癌旁正常组织,实时荧光定量PCR(qRT-PCR)检测两组miR-183表达差异。同时,qRT-PCR检测miR-183在非小细胞肺癌细胞系中的表达情况。通过转染技术,抑制miR-183表达。通过Transwell小室实验检测对非小细胞肺癌细胞侵袭、迁移的影响。通过Western Blot探索miR-183对非小细胞肺癌的作用机制。结果miR-183在非小细胞肺癌组织及癌细胞系中相比于正常组织或上皮细胞表达水平显著上升。Transwell小室实验结果显示,抑制miR-183表达后,非小细胞肺癌细胞侵袭、迁移能力受到抑制。Western Blot实验结果显示,在抑制miR-183表达后,上皮间质转化标志物蛋白N-cadherin、Vimentin、肿瘤干细胞标志物CD133、CD44表达显著下降。对Wnt/β-catenin信号通路关键蛋白检测,发现下调miR-183表达后,WNT1、p-β-catenin蛋白水平明显下降。结论miR-183在非小细胞肺癌中高表达,并且可能通过Wnt/β-catenin信号通路调控非小细胞肺癌上皮间质转化和肿瘤干细胞。 Objective To investigate the expression of miR-183 in non-small cell lung cancer and its underlying mechanism in influencing the disease.Methods Ten fresh non-small cell lung cancer tissues and adjacent normal tissues were collected.The differential expression of miR-183 between two groups was determined using quantitative real-time fluorescence PCR(qRT-PCR).The expression of miR-183 in non-small cell lung cancer cell lines was also evaluated by qRT-PCR.The expression of miR-183 was inhibited through transfection techniques.The impact on the invasion and migration of non-small cell lung cancer cells was assessed using Transwell assays.Furthermore,the role of miR-183 in non-small cell lung cancer was explored through Western Blot analysis.Results The expression levels of miR-183 were significantly elevated in non-small cell lung cancer tissues and cancer cell lines compared to normal tissues or epithelial cells.Inhibition of miR-183 expression resulted in the suppression of invasion and migration ability of non-small cell lung cancer cells,as observed in the Transwell assay.Western Blot analysis demonstrated a significant decrease in the expression of epithelial mesenchymal transformation marker proteins(N-cadherin,Vimentin)and tumor stem cell markers(CD133,CD44)following the inhibition of miR-183 expression.Additionally,the down-regulation of miR-183 expression led to a significant decrease in the levels of key proteins in the Wnt/β-catenin signaling pathway,including WNT1 and p-β-catenin.Conclusion miR-183 is highly expressed in non-small cell lung cancer and may play a regulatory role in epithelial mesenchymal transition and tumor stem cells through the Wnt/β-catenin signaling pathway.
