摘要
开发利用盐碱地种植大豆是提高我国大豆生产能力、保障粮食安全的重要途径,然而高盐碱环境严重抑制大豆生长,限制大豆产量。接种根瘤菌可增强大豆对盐胁迫的耐受性,从而提高盐碱地大豆产量。本研究从黄河三角洲黄河口地区分离到一株大豆根瘤菌DY23-9,经形态学观察和16S rRNA基因测序,鉴定该菌株为弗雷德中华根瘤菌(Sinorhizobium fredii)。DY23-9菌株可与11种耐盐碱大豆共生结瘤,能够在D-苹果酸、L-天冬酰胺酸、L-丝氨酸、D-甘露醇和N-乙酰基-D-葡萄胺等18种单一碳源条件下生长,最短世代时间仅为4.13 h。与模式菌株USDA110和USDA205相比,菌株DY23-9具有耐低温和耐盐优势。菌株DY23-9在实验室条件和大田条件下均对耐盐碱大豆TZX-805有显著的促生作用,并显著增加盐碱地大豆产量。以上结果表明土著根瘤菌DY23-9在盐碱环境中具有明显的生长优势,能够在黄河三角洲滨海盐碱地大豆生产中发挥促生增产作用。本研究为耐盐碱大豆根瘤菌的开发利用及盐碱地大豆高效种植技术的发展提供了菌株资源和理论依据。
Developing and utilizing saline-alkali land to cultivate soybean is an important way to improve soybean production capacity and ensure food security in China.However,high saline alkali environment se-verely inhibits soybean growth and yield.Inoculation of rhizobia can enhance the tolerance of soybean to salt stress,thereby increase the yield of soybean in saline-alkali land.In this study,a strain of soybean rhizobium DY23-9 was isolated from the coastal area of the Yellow River Delta.The strain was identified as Sinorhizobium fredii by morphology observation and 16S rRNA gene sequencing.The strain DY23-9 could grow symbiotically with 11 kinds of saline-tolerant soybean varieties,and could grow under 18 single carbon sources such as D-malic acid,L-asparagine acid,L-serine,D-mannitol and N-acetyl-D-glucosamine,and the shortest generation time was only 4.13 h.Compared with the model strains USDA110 and USDA205,the strain DY23-9 had the advantages of low temperature and salt tolerances.DY23-9 significantly promoted the growth of saline-tolerant soybean TZX-805 in both laboratory and field conditions,and significantly increased soybean yield in saline-alkali soil.These results indicated that the indigenous rhizobium DY23-9 had obvious growth advantages in sa-line-alkali environment,and could play roles in promoting growth and increasing production of soybean in the coastal saline-alkali lands of the Yellow River Delta.This study provided strain resource and theoretical basis for the development and utilization of saline-alkali tolerant soybean rhizobium and the development of efficient soybean planting technology in saline-alkali land.
