子宫主动脉结扎大鼠缺氧缺血脑病模型的声辐射力脉冲弹性成像结果与神经细胞凋亡率的相关性研究

摘要 目的:探讨子宫主动脉结扎大鼠缺氧缺血脑病模型的声辐射力脉冲弹性成像(acoustic radiation force impulse elastography,ARFI elastography)结果与神经细胞凋亡率的相关性。方法:选用30只孕晚期Wistar大鼠,通过子宫主动脉结扎的方法构建大鼠缺氧缺血性脑病(hypoxie-ischemic encephalopathy,HIE)模型。根据结扎时间分为结扎10分钟组、结扎20分钟组和对照组。每组模型在不同时间点利用彩色多普勒测定脑血流参数(Vs、Vd、RI),利用声辐射力脉冲弹性成像测定声触诊量化(VTQ)值。将HIE模型鼠部分大脑制作病理切片后进行HE染色和TUNEL细胞凋亡检测。最后通过统计分析计算超声结果和神经细胞凋亡率的相关性。结果:子宫动脉结扎10分钟组和20分钟组的脑血流参数Vs和RI值均低于对照组(P<0.05),而Vd值差异不显著(P>0.05)。两个HIE模型组的VTQ值高于对照组,且两组之间差异有统计学意义(P<0.05)。所有数值在各检测时间点之间没有明显变化。HE染色结果显示,HIE模型组脑部神经细胞发生明显变形。TUNEL实验显示模型组中脑神经细胞数目少于对照组,细胞凋亡率高于对照组。最后,通过计算得到,声辐射力脉冲弹性成像测量的VTQ值与脑神经细胞凋亡率呈显著高度相关性。结论:子宫主动脉结扎大鼠缺氧缺血脑病模型的声辐射力脉冲弹性成像结果与神经细胞凋亡率之间存在显著相关性。 Objective To investigate the correlation between the results of acoustic radiation force impulse elastography(ARFI elastography)and the rate of neuronal apoptosis in rat model of hypoxic-ischemic encephalopathy.Methods 30 Wistar rats in late pregnancy were used to construct a rat model of hypoxic-ischemic encephalopathy(HIE)by uterine aortic ligation.According to the ligation time,they were divided into control group,ligation group for 10 minutes and ligation group for 20 minutes.Cerebral blood flow parameters(Vs,Vd,RI)were measured by color Doppler ultrasound at different time points in each group,and virtual touch tissue quantification(VTQ)values were measured by ARFI elastography.HE staining and TUNEL cell apoptosis detection were performed after pathological sections were made in part of the brain of the model mice.Finally,the correlation between the ultrasound results and the apoptosis rate of nerve cells was calculated by statistical analysis.Results The values of cerebral blood flow parameters Vs and RI in the 10 min and 20 min uterine artery ligation groups were lower than those in the control group(P<0.05),while the difference in Vd values was not significant(P>0.05).The VTQ values of the two HIE model groups were higher than those of the control group,and the difference between the two groups was statistically significant(P<0.05).All the values did not change significantly between each detection time point.The results of HE staining showed that the neural cells in the HIE model group were obviously deformed.TUNEL assay showed that the number of brain neurons in the model group was less than that in the control group,and the apoptosis rate was higher than that in the control group.Finally,it was calculated that the VTQ values measured by ARFI elastography were highly correlated with the apoptosis rate of brain nerve cells.Conclusion There is a significant correlation between the results of acoustic radiation force impulse elastography and the apoptosis rate of nerve cells in the rat model of hypoxic-ischemic encephalopathy after uterine aorta ligation.
