丹参及其活性成分治疗血管性痴呆作用机制的研究进展 被引量:1

摘要 丹参其活性成分包括丹参酮ⅡA、丹参酸B、丹参素、隐丹参酮等。其中丹参酮ⅡA可通过抑制内质网应激抑制神经细胞凋亡,通过调节Toll样受体4(toll-like receptor 4,TLR4)/髓系分化初级反应蛋白88(myeloid differentiation primary response protein 88,MyD88)依赖性信号通路,下调β位淀粉样蛋白前体蛋白切割酶1(βamyloid precursor protein cleaving enzyme 1,BACE1)表达,调控核因子κB及其抑制蛋白的活性,减轻神经炎症反应,降低神经细胞炎性损伤;改善胆碱能功能对神经系统发挥保护作用;通过上调磷酸化-细胞外调节蛋白激酶(phosphorylation-extracellular regulated protein kinase, p-ERK)/细胞外调节蛋白激酶(extracellular regulated protein kinase, ERK)、蛋白激酶C受体1(protein kinase C receptor 1,RACK1)和抑制自噬来减轻神经元损伤。丹参酸B上调胰岛素样生长因子1(insulin-like growth factor 1,IGF-1)/蛋白激酶B(protein kinase B,Akt)信号通路、调节信号转导及转录激活蛋白3(transcription activating protein 3,STAT3)/促血管生长因子(vascular endothelial growth factor, VEGF)信号通路,抑制神经细胞凋亡,通过激活低密度脂蛋白受体相关蛋白6(low density lipoprotein receptor-associated protein 6,LRP6)/Wnt/β-连环蛋白(β-catenin)信号通路减轻神经元氧化应激损伤,还能通过抑制氧化应激反应促使突触蛋白恢复,以改善神经功能和认知功能。丹参素调节磷脂酰肌醇3激酶/Akt信号通路,抑制神经元凋亡,发挥神经保护作用。隐丹参酮可通过抑制脑血管内皮细胞中的β淀粉样蛋白聚集发挥抗血管性痴呆的作用。现将近年来丹参活性成分用于血管性痴呆的研究报道进行综述总结,为丹参的临床运用及血管性痴呆的治疗提供理论依据。 The active ingredients of Salvia miltiorrhiza include tanshinone ⅡA,tanshinone B,tanshinol,cryptotanshinone and so on.Tanshinone ⅡA can inhibit neuronal apoptosis by inhibiting endoplasmic reticulum stress.Tanshinone ⅡA regulates the Toll-like receptor 4(TLR4)/myeloid differentiation primary reactive protein 88(MyD88)-dependent signaling pathway,down-regulates the expression ofβ-site amyloid precursor protein cleaving enzyme 1(BACE1),and regulates the activities of NF-κB and IκB.It can reduce the neuroinflammatory response and reduce the inflammatory damage of nerve cells.It improves cholinergic function and protects the nervous system.It alleviates neuronal injury by up-regulating phospho-extracellular regulated protein kinase(p-ERK)/extracellular regulated protein kinase(ERK),protein kinase C receptor 1(RACK1)and inhibits autophagy.Salvianolic acid B up-regulates insulin-like growth factor 1(IGF-1)/protein kinase B(Akt)signaling pathway,regulates signal transducer and activator of transcription 3(STAT3)/pro-angiogenic growth factor(VEGF)signaling pathway,and inhibits neuronal apoptosis.It alleviates neuronal oxidative stress injury by activating the low-density lipoprotein receptor-associated protein 6(LRP6)/Wnt/β-catenin signaling pathway,and can also promotes the recovery of synaptic proteins by inhibiting oxidative stress response to improve neurological function and cognitive function.Danshansu plays a neuroprotective role by regulating PI3K/Akt signaling pathway and inhibiting neuronal apoptosis.Cryptotanshinone plays an anti-vascular dementia role by inhibiting the aggregation of Aβin cerebrovascular endothelial cells.This article reviews and summarizes the research reports of active ingredients of Salvia miltiorrhiza used in VaD in recent years,so as to provide theoretical basis for the clinical application of Salvia miltiorrhiza and the treatment of VaD.
