补肾调经汤对PCOS模型大鼠卵巢t-PA、PAI-1表达的影响

摘要 目的:观察补肾调经汤对PCOS模型大鼠卵巢t-PA、PAI-1表达的影响。方法:用来曲唑造模法建立雌性大鼠PCOS模型,设置正常对照组、模型组、西药组、中药组。通过阴道涂片观察大鼠动情周期,ELISA法检测大鼠血清T水平,电镜观察卵巢形态,Western blot检测测定大鼠卵巢t-PA、PAI-1蛋白的表达。结果:与正常对照组比较,模型组大鼠血清T水平升高,窦卵泡数增多,颗粒细胞层数减少,卵巢组织t-PA表达降低、PAI-1表达升高(P<0.01);与模型组比较,中药组和西药组血清T水平下降,窦卵泡数减少,颗粒细胞层数增多,卵巢组织t-PA表达升高、PAI-1表达降低(P<0.01)。结论:PCOS的发病可能与卵巢t-PA、PAI-1的异常表达有关,补肾调经汤可能能够通过调节t-PA、PAI-1的表达恢复卵巢正常生殖功能,维持生殖内分泌稳态,达到治疗PCOS的作用。 Objective:To observe the effect of Bushen Tiaojing Decoction on the expression of t-PA and PAI-1 in ovary of PCOS model rats.Methods:The PCOS model of female rats was established by using the method of trazol modeling,and the normal control group,model group,western medicine group and Chinese medicine group were set up.The estrous cycle of rats was observed by vaginal smear.The serum T level was detected by ELISA.The ovarian morphology was observed by electron microscope,and the expression of t-PA and PAI-1 protein in rat ovary was detected by Western blot.Results:Compared with the normal control group,the serum T level of rats in the model group increased,the number of sinus follicles increased,the number of granulosa cells decreased,and the t-PA expression in ovarian tissue decreased while the PAI-1 expression increased(P<0.01).Compared with the model group,the serum T level decreased,the number of sinus follicles decreased,the number of granulosa cells increased,and the t-PA expression in ovarian tissue increased while the PAI-1 expression decreased in the Chinese medicine group and the Western medicine group(P<0.01).Conclusion:The pathogenesis of PCOS may be related to the abnormal expression of t-PA and PAI-1 in ovary.Bushen Tiaojing Decoction may restore the normal reproductive function of ovary by regulating the expression of t-PA and PAI-1,maintain the reproductive endocrine homeostasis,and achieve the effect of treating PCOS.
