茯苓甘草汤对功能性消化不良模型大鼠胃组织及海马组织中AQP_(3)、AQP_(4)蛋白表达的影响 被引量:3

Effect of Fuling Gancao Decoction on the Expression of AQP3 and AQP4 Protein in Gastric Tissue and Hippocampus of Functional Dyspepsia Model Rats

摘要 目的:观察茯苓甘草汤对功能性消化不良(functional dyspepsia,FD)模型大鼠的胃组织的水液转运及脑部海马组织的影响。方法:72只SD大鼠随机分成正常组、模型组、茯苓甘草汤高剂量组、茯苓甘草汤中剂量组、茯苓甘草汤低剂量组、多潘立酮组。造模时用温度为0℃,0.5mol/L的稀盐酸(用量按0.8mL/100g)进行灌胃。正常组大鼠用等体积的自来水进行灌胃,每日1次,共造模14d。茯苓甘草汤高、中、低剂量组以及多潘立酮组分别进行给药灌胃,每日1次,连续7d。正常组及模型组在给药的7d内以等体积的生理盐水进行灌胃。结果:与正常组比较,模型组FD大鼠胃组织中水通道蛋白3(aquaporin 3,AQP_(3))、水通道蛋白4(aquaporin 4,AQP_(4))表达量降低(P<0.05);与模型组比较,茯苓甘草汤高、中、低剂量组及多潘立酮组胃组织中AQP_(3)、AQP_(4)表达量增加(P<0.05);与正常组比较,FD大鼠海马组织中AQP_(3)、AQP_(4)表达量降低(P<0.01),与模型组比较,茯苓甘草汤高、中、低剂量组及阳性组与模型组比较AQP_(3)、AQP_(4)表达量升高(P<0.01)。结论:茯苓甘草汤能通过促进胃组织内水液转运以及改变大鼠脑部海马组织中AQP_(3)、AQP_(4)的表达来改善FD的症状,可能是茯苓甘草汤治疗FD的作用机制之一。 Objective:To observe the effect of Fuling Gancao Decoction on transport of fluids in gastric tissue and hippocampus in functional dyspepsia(FD)model rats.Methods:Seventy-two SD rats were randomly divided into normal group,model group,high dose of Fuling Gancao Decoction group,middle dose of Fuling Gancao Decoction group,low dose of Fuling Gancao Decoction group,and domperidone group.During the modeling,0.5mol/L diluted hydrochloric acid(0.8mL/100g)at 0℃was used for intragastric administration.The rats in the normal group were intragastrically administered with equal volume of tap water once a day for 14 days.The high,medium and low dose groups of Fuling Gancao Decoction and the domperidone group were given intragastric administration once a day for 7 days.The normal group and the model group were intragastrically administered with an equal volume of normal saline within 7 days of administration.Results:Compared with the normal group,the expression of aquaporin 3(AQP3)and aquaporin 4(AQP4)in gastric tissue of FD rats in the model group decreased(P<0.05).Compared with the model group,the expression of AQP3 and AQP4 in the gastric tissue of the high,medium and low dose groups of Fuling Gancao Decoction and the domperidone group increased(P<0.05).Compared with the normal group,the expression of AQP3 and AQP4 in the hippocampus of FD rats decreased(P<0.01).Compared with the model group,the expression of AQP3 and AQP4 in the high,medium and low dose groups of Fuling Gancao Decoction and the positive group increased compared with the model group(P<0.01).Conclusion:Fuling Gancao Decoction can improve the symptoms of FD by promoting transport of fluids in gastric tissue and changing the expression of AQP3 and AQP4 in hippocampus of rats,which may be one of the mechanisms of Fuling Gancao Decoction in the treatment of FD.
