摘要
目的:探讨聚嘧啶束结合蛋白2(polypyrimidine tract binding protein 2,PTBP2)在肌萎缩侧索硬化症(amyotrophic lateral sclerosis,ALS)发展中的作用及相关机制。方法:采用免疫荧光染色法和Western Blot分析检测野生型(wild type,WT)和转基因(transgenic,TG)(SOD1*G93A)小鼠脊髓中PTBP2的表达和分布,分析PTBP2表达和分布与神经元细胞死亡之间的可能联系。结果:在ALS的发病前、发病和进展阶段,PTBP2在前角、后角和中央管周围区域的表达和分布显著增加。星形胶质细胞和小胶质细胞以及成体神经元表达PTBP2蛋白。ALS相关的神经元细胞死亡与PTBP2表达和分布的增加密切相关。表达PTBP2的神经元细胞在ALS进展过程中丢失。结论:PTBP2在脊髓中的表达和分布变化可能与ALS的病因有关,PTBP2表达的增加与ALS中神经元细胞的丢失相关。
Objective:To explore the role and related mechanisms of polypyrimidine tract binding protein 2(PTBP2)in the development of amyotrophic lateral sclerosis(ALS).Methods:Immunofluorescence and Western Blot analysis were used to detect the expression and distribution of PTBP2 in the spinal cord of wild-type(WT)and transgenic(TG)(SOD1*G93A)mice,and to analyze the possible relationship between PTBP2 expression and distribution and neuronal cell death.Results:The expression and distribution of PTBP2 in the anterior horn,posterior horn,and periventricular region increased significantly during the pre-onset,onset,and progression stages of ALS.Astrocytes,microglia,and adult neurons expressed the PTBP2 protein.The neuronal cell death associated with ALS was closely related to the increase in PTBP2 expression and distribution.Neuronal cells expressing PTBP2 were lost during the progression of ALS.Conclusion:The changes in the expression and distribution of PTBP2 in the spinal cord may be related to the pathogenesis of ALS,and the increase in PTBP2 expression is associated with the loss of neuronal cells in ALS.
