摘要
试验旨在探究不同品种乳鸽肌纤维特征的差异和分子调控机制。试验以23日龄银王鸽、石岐鸽和欧洲肉鸽乳鸽的胸肌为研究对象,通过HE染色观察胸肌肌纤维表型差异,并通过转录组测序筛选可能影响不同鸽品种胸肌发育差异的基因,进行实时荧光定量qPCR验证。结果显示:三个品种乳鸽活体重、屠体重、屠宰率、肌纤维直径和密度在品种和性别间均存在较大差异;在不同品种的公鸽胸肌转录组数据中筛选出与肌肉发育和脂肪代谢相关的99个差异表达基因(DEGs),发现DEGs主要参与肌肉收缩和脂肪酸氧化代谢过程,并构建了5个基因互作网络。结果表明,不同品种不同性别乳鸽肌纤维特征存在较大差异,而筛选到的不同品种、性别中差异表达的成肌和成脂基因将为今后进一步开展肉鸽生长性能分子标记辅助育种提供数据基础。
The purpose of this study was to explore the differences in myofiber characteristics and molecular regulation mechanism among different pigeon breeds.In this study,breast muscle of 23-day-age Silver king pigeon,Sheek-kee pigeon and European meat pigeon squabs were taken as objects.The phenotypic differences of breast muscle myofibers were observed by HE staining,and genes that may affect the breast muscle development of different pigeon breeds were screened by transcriptome sequencing,and verified by real-time fluorescent quantitative qPCR.It was found that the live weight,slaughter weight,slaughter rate,myofiber diameter and density of the three pigeon breeds were significantly different between breeds and sexes;Ninety-nine differentially expressed genes(DGEs)related to myogenesis and adipogenesis were identified.It was found that DEGs were mainly involved in muscle contraction and fatty acid oxidation metabolism biological processes,and five gene interaction networks were built.The results indicated that there were significant differences in myofiber characteristics among different breeds and gender of pigeon squabs,and the differential expression of myogenesis and adipogenesis related genes among different breeds and genders would provide data basis for molecular marker-assisted breeding of growth performance of meat pigeons in the future.
