摘要
目的:探讨基质金属蛋白酶3(Matrixmetalloproteinase3,MMP-3)11715A/6A基因多态性与新疆维吾尔族成人慢性牙周炎的相关性。方法:从前期流行调查组获取的墨玉县样本库中随机抽取300例颊黏膜拭子样本(分为健康对照组、轻度组以及中重度组三组,每组100例),提取DNA并采用限制性片段长度多态性聚合酶链反应检测MMP-311715A/6A位点的基因型分布,使用卡方检验、Kruskal-Wallis H检验、Mann-Whitney U检验以及多分类Logistic回归分析MMP-311715A/6A基因多态性与牙周炎易感性的相关性。结果:MMP-311715A/6A的5A6A和6A6A基因型分布在健康对照组与轻度组之间、健康对照组与中重度组间比较,差异均具有统计学意义(P<0.05);5A6A等位基因分布健康对照组与轻度组间的差异具有统计学意义(P<0.05)。多分类Logistic回归分析显示,年龄<35岁、5A/6A基因型及5A等位基因是慢性牙周炎的保护因素(P<0.05)。结论:MMP-311715A/6A基因多态性可能与新疆墨玉县维吾尔族人群慢性牙周炎相关,但仍需要更高质量的多中心研究进一步证实。
Objective To investigate the correlation between matrix metalloproteinase 311715A/6A polymorphisms and adult chronic periodontitis among Uyghur adults in Xinjiang.Methods 300 buccal mucosal swab samples were randomly selected from the Moyu County sample base obtained from the previous epidemiological investigation group.DNA was extracted and polymerase chain reaction restricted fragment length polymorphism was used to detect the genotype distribution of MMP-311715A/6A.Chi-square test,Kruskal-Wallis H test,Mann-Whitney U test and multivariate Logistic regression were used to analyze the correlation between MMP-31171 polymorphism and periodontitis susceptibility.Results The case-control study showed that there were significant difierences in 5A6A and 6A6A genotype distribution between healthy controls and mild periodontitis and between mild periodontitis and moderate to severe periodontitis(P<0.05).There was also significant difierence in allele frequency between healthy controls and mild periodontitis(P<0.05).Logistic analysis showed that age under 35 years old,5A/6A genotype and 5A allele were the protective factors of chronic periodontitis(P<0.05).Conclusion The polymorphism of MMP-311715A/6A may be related to chronic periodontitis among Uyghur adults in Moyu County of Xinjiang,but higher quality multi-center studies are still needed for further confirmation.
