摘要
【目的】获取独脚金[Striga asiatica(L.)O. Kuntze.]的转录组信息特征。【方法】提取独脚金叶片和茎的总RNA、建库、测序、组装,以注释和比对等生物信息分析技术得到独脚金转录组的遗传信息。【结果】共获得38 096 466个高质量Reads,通过de novo组装获得41 918条Unigenes,N50长度为1 754 bp,平均长度1 029 bp。共有27 375条Unigenes(占65.31%)在CDD、PFAM、KEGG、KOG、GO、NR、NT等数据库中得到注释。其中NR数据库注释到27 187条Unigenes,在KEGG中注释到5 331条Unigenes,涉及284条代谢通路。在独脚金中共鉴定到30条Unigenes涉及黄酮类生物合成,同时还鉴定到778个转录因子(TFs)。采用微卫星识别软件(MISA)对组装的1 000 bp以上的Unigenes分析发现,13 107条Unigenes包含17 036个简单重复序列(SSR)位点。【结论】利用转录组数据分析为独脚金的重要功能基因鉴定和次生代谢通路的挖掘奠定了研究基础。
Objective To obtain a transcriptome information profile of Striga asiatica(L.)O.Kuntze..Methods The total RNA from leaves and stems was extracted,the database was library constructed,high-throughput sequenced and assembled,and the genetic information of the transcriptome of Striga asiatica(L.)O.Kuntze.was obtained by annotation and bioinformatics analysis.Results A total of 38096466 high-quality Reads were obtained,and 41918 unigenes were obtained by de novo assembly,with an N50 length of 1754 bp and an average length of 1029 bp.A total of 27375 unigenes(65.31%)were annotated in CDD,PFAM,KEGG,KOG,GO,NR,NT and other databases.Among them,27187 unigenes were annotated in NR database and 5331 unigenes were annotated in KEGG,involving 284 metabolic pathways.A total of 30 unigenes involving flavonoid biosynthesis and 778 transcription factors(TFs)were also identified.Analysis of assembled unigenes over 1000 bp using microsatellite identification tool(MISA)revealed that 13107 unigenes contained 17036 simple sequence repeat(SSR)locis.Conclusion Transcriptome data analysis was used to lay the foundation of research for the identification of important functional genes and mining of secondary metabolic pathways in Striga asiatica(L.)O.Kuntze..
