摘要
目的:观察苗药白龙须对类风湿关节炎患者滑膜成纤维细胞(RA-FLS)增殖及STAT-3相关炎症通路的影响,揭示白龙须对RA滑膜炎症的调控机制。方法:培养3~5代RA-FLS细胞,分为空白组(胎牛血清)、正常组(正常兔血清)、雷公藤多苷(TG)组、来氟米特(LEF)组、白龙须低剂量组、白龙须中剂量组和白龙须高剂量组,按各组剂量加含药血清培养,用CCK8法检测各组RA-FLS增殖抑制率的差异,ELISA法检测RA-FLS与CD4+T细胞共培养上清液中炎症细胞因子IL-6、IL-17、IL-23和TNF-α浓度,蛋白印迹法(Wersten Blots)检测RA-FLS中pSTAT-3、SOCS-3的蛋白表达。结果:白龙须能够抑制RA-FLS的增殖,降低共培养体系细胞上清中IL-6、IL-17、IL-23、TNF-α等4种炎症细胞因子的浓度,降低RA-FLS中pSTAT-3的表达量,同时上调SOCS-3蛋白表达。结论:白龙须可能通过抑制STAT-3/IL-17炎症通路的活化发挥抗炎作用,抑制RA-FLS持续性增殖。
Objective To observe the effect of Miao medicine Bailongxu on the proliferation of synovial fibroblasts(RA-FLS)and the inflammatory pathway related to STAT-3 in patients with rheumatoid arthritis,and to reveal the regulation mechanism of the inflammatory pathway related STAT-3/IL-17 in synoviocytes.Method We selected 3-5 generations of RA-FLS cells,and divided them into blank group(fetal calf serum),normal group(normal rabbit serum),triptolide(TG)group,leflunomide(LEF)group,Bailongxu low-dose group,Bailongxu medium-dose group and Bailongxu high-dose group.The cultures were cultured according to the prescribed dose with serum.Differences in proliferation inhibition of RA-FLS in each groups were determined by CCK8.The concentrations of inflammatory cytokines IL-6,IL-17,IL-23,and TNF-αwere determined in the supernatants jointly containing RA-FLS with CD4+T cells by ELISA.The protein expression of pSTAT-3 and SOCS-3 in RA-FLS was detected by the protein blotting method(Wersten Blots).Results It could inhibit the proliferation of RA-FLS,reduce the concentration of four inflammatory cytokines:IL-6,IL-17,IL-23,and cell-TNF-α,and reduce pSTAT-3 expression and upregulate the SOCS-3 protein expression in RA-FLS.Conclusion It may exert an anti-inflammatory effect and inhibit RA-FLS persistent proliferation by inhibiting the activation of STAT-3/IL-17 inflammatory pathway.
