平喘颗粒调控M2型巨噬细胞极化改善哮喘小鼠气道炎症及气道重塑的实验研究 被引量:1

摘要 目的 观察平喘颗粒对哮喘小鼠气道炎症、气道重塑及M2型巨噬细胞极化相关因子的影响。方法 选择30只6~8周雌性BALB/C小鼠随机分为空白组、模型组、平喘颗粒组,采用卵清蛋白和氢氧化铝构建哮喘小鼠模型,给予平喘颗粒进行干预。采用HE和PAS染色检测各组小鼠肺组织病理形态变化;吉姆萨染色计数各组小鼠肺泡灌洗液中总细胞数、嗜酸性粒细胞数、中性粒细胞数、巨噬细胞数;ELISA法检测各组小鼠血清IgE含量及肺泡灌洗液中巨噬细胞M2标记物Arg1、白细胞介素-10(IL-10)的含量;real-time PCR检测各组小鼠肺组织中巨噬细胞M2标记物YM1、MRC1的水平;免疫荧光检测肺组织F4/80与MRC1的表达与共定位。结果 与模型组相比,平喘颗粒组小鼠肺组织炎性细胞浸润情况明显减少,气道黏液及杯状细胞明显减轻。与模型组相比,平喘颗粒组BALF中白细胞总数、嗜酸性粒细胞数量、中性粒细胞数量、巨噬细胞数量明显降低,差异有统计学意义(P <0.05)。平喘颗粒组小鼠血清中IgE水平明显低于模型组,但高于空白组,与模型组及空白组比较差异均有统计学意义(P <0.05)。与模型组相比,平喘颗粒组小鼠BALF中Arg1和IL-10水平明显降低(P <0.05),与空白组比较无明显差异(P> 0.05);平喘颗粒组小鼠肺组织中YM1、MRC1基因表达水平明显降低,比较差异有统计学意义(P <0.05),与空白组比较无明显差异(P> 0.05)。免疫荧光检测各组小鼠肺组织结果显示MRC1与F4/80的表达与共定位荧光强度明显降低。结论 平喘颗粒可以抑制哮喘小鼠气道炎症及气道重塑,其机制可能与本方能够抑制哮喘小鼠M2型巨噬细胞极化的相关因子有关。 Objective:To observetheeffectsof Pingchuan Granuleson airway inflammation,airway remodeling and related factors of M2-type macrophage polarization in asthmatic mice.Methods:30 female BALB/C mice at 6~8 weeks were randomly divided into blank group,model group and Pingchuan Granules group.The mice model of asthma was constructed using obumin and aluminum hydroxide.The mice were given Pingchuan Granules for in-tervention.Jimsa staining was used to count the total cell count,eosinophil count,neutrophil count and macro-phage count in alveolar lavage fluid of each group.Serum IgE content of each group was detected by ELISA.The contents of macrophage M2 markers Argl and IL-10 in alveolar lavage fluid of mice in each group were deter-mined by ELISA.The levels of macrophage M2 markers YM1 and MRC1 in lung tissues of mice in each group were detected by real-time PCR.The expression and co-localization of F4/80 and MRC1 in lung tissues were de-tected by immunofluorescence.Results:Compared with model group,the inflammatory cell infiltration in lung tis-sue,airway mucus and goblet cells were significantly reduced in Pingchuan Granules group.Compared with the model group,the total number of white blood cells,the number of eosinophils,the number of neutrophils and the number of macrophages in BALF of Pingchuan Granules group were significantly decreased,with statistical differ-ence(P<0.05).Serum IgE level in Pingchuan Granules group was significantly lower than that in model group,but higher than that in blank group,which both had statistical difference compared with model group and blank group(P<0.05).Compared with model group,Argl and IL-10 levels in BALF of mice in Pingchuan Granules group were significantly decreased,with statistical difference(P<0.05),but showed no statistical difference com-pared with blank group(P>0.05).The expression levels of YM1 and MRC1 genes in lung tissues of mice in Ping-chuan Granules group were significantly decreased,with statistical difference(P<0.05),but showed no statistical difference compared with blank group(P>0.05).Immunofluorescence detection of the expression and co-localiza-tion of F4/80 and MRC1 in lung tissues of mice in each group showed that the distribution of MRC1 expression in Pingchuan Granules group was significantly reduced.Conclusion:Pingchuan Granules can inhibit airway inflam-mation and remodeling in asthmatic mice,and the mechanism may be related to the related factors of inhibiting M2-type macrophage polarization in asthmatic mice.
