摘要
制糖业是云南的主要产业之一,每年会产生大量的蔗渣.为了能有效地处理云南制糖业产生的蔗渣,并对其进行再次利用,研究团队从云南省昆明市筇竹寺森林公园采集土壤样品,分别用CMC刚果红培养基和以蔗渣作为唯一碳源的蔗渣培养基进行纤维素分解细菌的筛选,通过测定其在2种培养基上形成的水解透明圈和菌落直径的比值进行分析,再结合纤维素酶活力变化特征筛选获得一株能高效分解利用蔗渣纤维素的细菌QZS⁃1.该菌株整体为乳白色,边缘呈不规则锯齿状,上有花纹.经过革兰氏染色和芽孢染色确定为革兰氏阳性的芽孢杆菌.同源性比对显示,其与短小芽孢杆菌同源性达99.91%,最终鉴定为短小芽孢杆菌(Bacillus pumilus),在72 h时产酶活力达到最高,为17.217 U/mL.
Yunnan Province,as a major province having sugar industries,produces a large amount of bagasse and hence it is urgent to address the issues related to bagasse accumulation.The cellulose⁃decomposing strains were isolated from the soil obtained from Qiongzhusi Forest Park in Kunming of Yunnan Province,and were screened using carboxymethyl cellulose Congo red and bagasse mediums as the only carbon sources.The ratio of transparent circle and colony diameter formed on the two mediums was determined.Bacillus pumilus was identified as the high⁃efficiency cellulase⁃producing strains QZS⁃1.This strain was milky white overall,with irregular serrated edges and patterns.The Bacillus was identified as Gram⁃positive by Gram staining and spore staining.After homology comparison,it shared 99.91%homology with Bacillus pumilus,and was finally identified as Bacillus pumilus.It reached the highest enzyme production activity after 72 hours(17.217 U/mL).
