基于多光子显微成像的隆突性皮肤纤维肉瘤定量分析

摘要 多光子显微成像是一种新型的无创诊断技术。与传统的诊断方法相比,多光子显微镜具有穿透深度更深且光毒性和光漂白较低等优势,在皮肤病学领域具有广阔的应用前景。本研究的目的是通过利用多光子显微成像技术对隆突性皮肤纤维肉瘤的未染色组织切片进行成像,试图寻找一种有效的无损成像方法,替代传统的基于苏木精和伊红(H&E)染色的病理诊断,同时提高DFSP诊断的准确性。结果表明,利用多光子显微镜对无H&E染色的组织切片进行成像,可以识别DFSP表皮和真皮中的细胞和基质以及皮下组织的组织学形态,结果与传统组织学分析表征类似。 Multiphoton microscopy(MPM)is a new non-invasive diagnostic technology.Compared with traditional diagnosis methods,Multiphoton microscopy has the advantages of deeper penetration,lower phototoxicity and photobleaching.It has broad application prospects in the field of dermatology.The purpose of this study was to find an effective non-destructive imaging method by using the multiphoton microscope to image the unstained tissue sections of dermatofibrosarcoma protuberans(DFSP),replace the traditional pathological diagnosis based on hematoxylin and eosin(H&E)staining while improving the accuracy of diagnosing DFSP.The results show the patterns of DFSP extending into the subcutaneous tissue and histology procedures of cancer cells and matrix can be identified by imaging the tissue sections without H&E staining with multiphoton microscope.The results were similar to the traditional histological analysis.
