摘要
采用超声波辅助双水相提取花生壳总黄酮,以黄酮得率为指标,在单因素实验的基础上,通过响应面分析法优化提取工艺,并研究其抗氧化活性。实验结果表明,黄酮提取的最佳工艺参数为:浸提温度52℃,投料量60 mg,乙醇质量分数30%,磷酸氢二钾质量分数21%,超声时间20 min。在此条件下,黄酮得率预测值为4.31%,验证值为4.30%,与预测值接近,因此认为回归模型有效。抗氧化结果表明,花生壳总黄酮对DPPH自由基、羟基自由基和超氧阴离子自由基的清除率分别为92.33%、57.89%和97.15%,表现出良好的抗氧化活性。
Ultrasonic assisted aqueous two-phase extraction was used to extract total flavonoids from peanut shells.Taking the yield of flavonoids as index and based on the single factor experiments,the extraction process was optimized by response surface analysis,and the antioxidant activity of the flavonoids was also studied.The experimental results showed that the optimal technological parameters for the extraction of flavonoids were as follows:extraction temperature 52℃,feeding amount 60 mg,ethanol mass fraction 30%,potassium hydrogen phosphate mass fraction 21%,ultrasonic time 20 min.Under these conditions,the predicted value of flavonoids yield was 4.31%,and the verified value was 4.30%,which were close to each other,so the regression model was considered to be effective.The antioxidant results showed that the scavenging rates of total flavonoids from peanut shells to DPPH radicals,hydroxyl radicals and superoxide anion radicals were 92.33%,57.89%and 97.15%,respectively,exhibiting good antioxidant activity.
