LncRNA TMPO-AS1通过PI3K/Akt信号通路对多囊卵巢综合征大鼠性激素和卵巢功能的影响 被引量:1

Effects of LncRNA TMPO-AS1 on sex hormones and ovarian function in rats with polycystic ovary syndrome through PI3K/Akt signaling pathway

摘要 目的 探究LncRNA TMPO-AS1对多囊卵巢综合征(PCOS)大鼠的影响以及其对磷脂酰肌醇-3-激酶(PI3K)/蛋白激酶B(Akt)通路相关分子的调控作用。方法 选取36只SPF级SD雌性大鼠,按照随机数字表法分为模型组(A组)、空白质粒组(B组)、TMPO-AS1过表达质粒组(C组)共3组,每组12只大鼠。采用来曲唑注射法制备大鼠PCOS模型,于造模第1天起,B、C组大鼠尾静脉注射空白质粒及TMPO-AS1过表达质粒,A组注射等量0.9%氯化钠溶液,连续注射21 d。比较各组大鼠的性激素[促黄体生成素(LH)、睾酮、卵泡刺激素(FSH)]水平、卵巢功能(卵巢重量/体重比例、成熟卵泡数量、黄体数量)、卵巢组织的PI3K、Akt mRNA和蛋白表达量以及子宫容受性[白血病抑制因子(LIF)蛋白和整合素β3(ITG β3)]、代谢功能[空腹血糖(FBG)、空腹胰岛素(FINS)、HOMA-IR]水平。结果 C组大鼠的性激素LH、睾酮水平、卵巢重量/体重比例、成熟卵泡数量、FBG、FINS、HOMA-IR水平均高于A、B组,FSH水平、黄体数量均低于A、B组,同时C组大鼠的PI3K、Akt mRNA及蛋白表达量、LIF、ITG β3水平均低于A、B组,差异均有统计学意义(P<0.05);但A组、B组间各项指标比较,差异均无统计学意义(P>0.05)。结论 LncRNA TMPO-AS1能够加重PCOS大鼠性激素水平和代谢功能紊乱和卵巢功能受损,降低子宫容受性,可能与下调PI3K、Akt mRNA和蛋白表达有关。 Objective To investigate the effect of LncRNA TMPO-AS1 on polycystic ovary syndrome(PCOS)rats and its regulation on phosphatidylinositol-3-kinase(PI3K)/protein kinase B(Akt)pathway related molecules.Methods Thirty-six SPF SD famale rats were divided into model group(group A),blank plasmid group(group B)and TMPO-AS1 overexpression plasmid group(group C)according to the random number table,with 12 rats in each group.The PCOS rat model was prepared by letrozole injection.The rats in group B and C were injected with blank plasmid and TMPO-AS1 overexpression plasmid in the tail vein,and the rats in group A were injected with the same amount of normal saline.The injections continued for 21 days.Sex hormone[luteinizing hormone(LH),testosterone,follicle stimulating hormone(FSH)]levels,ovarian function(ovarian weight/body weight ratio,number of mature follicles,number of corpus luteum),PI3K mRNA expression levels and PI3K,Akt protein expression levels in ovarian tissue,uterine receptivity[leukemia inhibitory factor(LIF)protein and integrinβ3(ITGβ3)]and metabolic function[fasting blood sugar(FBG),fasting insulin(FINS),HOMA-IR]were compared among all groups.Results The levels of sex hormones LH,testosterone and ovarian weight/weight to mass ratio of sex hormones and the number of mature follicles,levels of FBG,FINS,HOMA-IR in group C were higher than those in groups A and B,but the levels of FSH and the number of corpus luteum in group C were lower than those in groups A and B,at the same time,the expression levels of PI3K,Akt mRNA and protein,ILF and ITGβ3 protein in group C were lower than those in groups A and B,the differences were statistically significant(P<0.05).There were no statistically significant differences in various indicators between group A and group B(P>0.05).Conclusion LncRNA TMPO-AS1 can aggravate the disturbance of sex hormone levels and metabolic function,and impaired ovarian function in PCOS rats,decrease the uterine receptivity,which may be related to the down-regulation of PI3K,Akt mRNA and protein expressions.
