摘要
【目的】比较6种不同人参属药材提取物的体内外抗炎活性差异,并探讨其抗炎活性机制。【方法】采用CCK-8法检测RAW264.7细胞活力,Griess法检测NO释放,ELISA法检测细胞分泌炎症因子情况,Western blot法检测NF-κB信号通路中关键蛋白表达情况。采用二甲苯所致小鼠耳肿胀模型,检测耳肿胀度及HE染色的病理切片情况。【结果】在给药浓度为0.02、0.20 mg/mL时,6种人参属药材提取物对RAW264.7细胞活力无显著抑制作用(P>0.05)。6种人参属药材提取物在0.2 mg/mL浓度下均显著抑制LPS诱导的RAW264.7细胞上清液中NO含量的升高(P<0.05或P<0.01或P<0.001),其中屏边三七抑制作用最强。与LPS组相比,6种人参属药材提取物均显著降低细胞分泌的TNF-α含量(P<0.01或P<0.001)。屏边三七组和竹节参组均显著逆转LPS诱导的细胞中p-NF-κB、IκBα蛋白表达量的升高;而珠子参组和竹节参组均显著抑制NF-κB蛋白表达。此外,6种人参属药材提取物对二甲苯所致小鼠耳肿胀具有剂量依赖性抑制作用,其中屏边三七组、三七组、西洋参组及竹节参组对小鼠耳肿胀的抑制率分别为91.11%、68.89%、82.22%、64.44%。小鼠耳组织HE染色结果表明,模型组小鼠耳组织出现明显结缔组织水肿,间隙增加及炎性细胞浸润。6种人参属药材提取物组与模型组相比水肿反应和炎性细胞浸润减少,其中屏边三七改善效果最好。【结论】6种人参属药材具有体内外抗炎作用,且活性存在差异,其机制作用可能与调节NF-κB信号通路相关。
[Objective]The study aimed to compare the anti-inflammatory activities of six extracts from Panax species in vitro and in vivo,and to explore the anti-inflammatory mechanism.[Method]The viability of RAW264.7 cells was detected by CCK-8 method.NO release was detected by Griess method.The secretion of inflammatory factors was detected by ELISA.The expressions of key proteins were detected by Western blot.[Result]At concentrations of 0.02 and 0.20 mg/mL,six extracts from Panax species showed no significant inhibitory effect on RAW264.7 cell viability(P>0.05).Six extracts significantly inhibited the increase of NO content in the supernatant of RAW264.7 cells induced by LPS at a concentration of 0.2 mg/mL(P<0.05 or P<0.01 or P<0.001),with Panax stipuleanatus having the strongest in-hibitory effect.Compared with LPS group,six extracts significantly reduced TNF-αcontent in cells(P<0.01 or P<0.001).P.stipuleana-tus group and P.japonicus group demonstrated obviously down-regulated p-NF-kB and IkBαprotein expressions,while P.japlcus group and P japonicus group significantly inhibited NF-B protein expression.In addition,six extracts showed dose-dependent inhibitory ffect on xy-lene induced ear swling in mice.The inhibitory rates of P.stipuleanatus group,P.notoginseng group,P.quinquefolius group,and P.japonicus group on mouse ear swelling were 91.11%,68.89%,82.22%,and 64.44%,respectively.The HE staining results of mouse ear tissue showed that the model group showed significant connective tissue edema,increased gaps,and inflammatory cell infiltration in the ear tissue.Compared with the model group,six extracts reduced edema reactions and inflammatory cell infiltration,with P.stipuleanatus group having the best improvement effect.[Conclusion]Six Panax species have anti-inflammatory effects in vivo and in vitro,and there are differences in anti-inflammatory activities.The mechanism may be related to the regulation of NF-kB signaling pathway.
