摘要
试验以黑曲霉固态发酵葡萄皮渣为研究对象,以木聚糖酶的活性为指标,应用响应面法对产木聚糖酶的工艺进行优化。首先对5个因素进行单因素试验,通过Plackett-Burman设计确定了3个显著性因素,分别是发酵时长、黑曲霉接种量和初始pH。经过Box-Behnken设计试验,建立二次回归模型,得到最佳发酵参数。结果表明,当初始pH为7、发酵时长为58 h、接种量为10%、初始含水量为60%、料层厚度为2.5 cm时,木聚糖酶最高活性为246.98 U/g,相较于优化前(88.62 U/g)提高了179%。由此可见,通过优化黑曲霉固态发酵葡萄皮渣的工艺条件可显著提高木聚糖酶活性。
The experiment focuses on the solid-state fermentation of grape skin residue by Aspergillus ni⁃ger,and the activity of xylanase is used as an indicator to optimize the production process of xylanase us⁃ing response surface methodology.Firstly,a single factor experiment was conducted on five factors,and three significant factors were determined through the Plackett-Burman design,namely fermentation dura⁃tion,inoculation amount of Aspergillus niger,and initial pH.After Box-Behnken design experiments,a quadratic regression model was established to obtain the optimal fermentation parameters.The results showed that when the initial pH was 7,the fermentation time was 58 hours,the inoculation amount was 10%,the initial water content was 60%,and the material layer thickness was 2.5cm,the highest activity of xylanase was 246.98 U/g,which was 179%higher than the optimized(88.62 U/g).It can be seen that optimizing the process conditions for solid-state fermentation of grape skin residue by Aspergillus niger can significantly improve the activity of xylanase.
