‘桂粉葛1号’组织培养再生体系的建立 被引量:1

摘要 目的:粉葛传统的栽培方式主要有压条繁殖、块茎苗、扦插繁殖等,繁殖效率较低,为建立粉葛组培再生体系,实现粉葛组培苗的工厂化生产。方法:本文以‘桂粉葛1号’嫩茎段为外植体,研究了灭菌时间、植物生长调节剂配比、移栽基质配比对‘桂粉葛1号’组织培养过程中外植体消毒、启动培养、继代培养、生根培养及组培苗移栽的影响。结果:实验结果如下:以75%酒精30 s+0.1%氯化汞8 min组合进行消毒,外植体成活率达83.45%;启动及继代培养基为MS+0.4 mg/L 6-BA+0.1 mg/L NAA,腋芽萌芽率达92.22%,继代增殖系数约2.61;生根培养基为MS+0.3 mg/L NAA,同时增加蔗糖添加浓度至30g/L,可明显提高组培苗根长、根数及根粗等指标,生根率达95.00%;组培苗移栽基质为黄泥:珍珠岩:营养基质(1∶1∶1),移栽存活率达98.00%。结论:本研究初步构建了高效的‘桂粉葛1号’组培再生体系,为粉葛组培营养杯苗的工厂化生产提供参考依据。 Pueraria lobata,a plant commonly propagated through traditional methods such as layering,tuber seedlings,and cuttings,exhibits low propagation efficiency.To address this issue and enable large-scale production of Pueraria lobata tissue culture seedlings,a tissue culture regeneration system was established.The effects of sterilization time,plant growth regulators and matrix ratio on disinfection,priming culture,subculture,rooting culture and tissue culture seedling transplanting of‘Guifenge No.1’were studied by tissues culture.With axillary bud as explants,its stem was used to analyze the costs oftissue culture seedling production at the two stages.The results showed that the explants were sterilized in 75%alcohol and 0.1%HgCl_(2) for 30 s and 8 min,the survival rate reached to 83.45%.The medium for the priming culture and subculture were MS+0.4 mg/L 6-BA+0.1 mg/L NAA,the percentage of axillary bud was up to 92.22%and multiplication coefficient reached to 2.61.The rooting medium was MS+0.3 mg/L NAA,with rooting rate of 95%.The increase of the sugar concentration to 30 g/L improved root quality such as root length,root number and root thickness.The transplanting substrate of tissue culture seedlings was yellow mud,perlite,and substrate,the ratio was 1:1:1 and the survival rate of transplanting was 98%.In this study,an efficient tissue culture regeneration system of'Guifengi No.1'was preliminarily constructed,providing a reference for the factory production of Pueraria lobata tissue culture nutrient cup seedlings.
