摘要
目的:探究电子加速器辐照灭菌对广枣药材和广枣水提液中有效成分没食子酸和总黄酮及微生物水平的影响。方法:采用2、4、6、8、10kGy五个辐照剂量对广枣去核粉末和广枣水提液进行电子加速器辐照,分别比较辐照前后的性状特征、有效成分没食子酸和总黄酮含量以及细菌菌落总数、霉菌、酵母菌菌落总数的变化。结果:不同剂量电子束辐照后,广枣去核药材粉末图谱相似度高达0.999及以上,没食子酸和总黄酮含量无显著性差异,细菌菌落数、霉菌酵母菌菌落总数显著性降低(P<0.05),当辐照剂量为6kGy时,微生物水平符合《中华人民共和国药典》2020版规定。广枣水提液性状、总黄酮未见明显差异,没食子酸含量显著降低(P<0.05),细菌菌落数、霉菌酵母菌菌落总数显著性降低(P<0.05),当辐照剂量为6kGy时,微生物水平符合《中华人民共和国药典》2020版规定。结论:不同剂量的电子束辐照灭菌对广枣去核药材质量无显著影响,且辐照剂量达6kGy时达到良好的灭菌效果。电子加速器辐照适用于广枣去核药材粉末灭菌,而对广枣水提液的质量影响显著,主要体现于降低没食子酸的含量。
Objective:To investigate the effect of electron accelerator irradiation sterilization on gallic acid,total flavone and microorganism levels in Fruotus choerospondiatis medicinal materials and its aqueous extract.Methods:Five irradiation doses of 2,4,6,8,10kGy were used to irradiate the seedless powder and water extract of Fruotus choerospondiatis with electron accelerator.The characteristics,the content of gallic acid and total flavone,the total bacterial colony,mold and yeast colony before and after irradiation were compared.Results:After electron beam irradiation at different doses,the similarity of the powder atlas of the seedless medicinal material of Fruotus choerospondiatis was as high as 0.999and above.There was no significant difference in the contents of gallic acid and total flavonoids,but the number of bacterial colonies and the total number of fungal yeast colonies decreased significantly(P<0.05).When the irradiation dose was 6kGy,the microbial level met the requirements of the Chinese Pharmacopoeia2020.There was no significant difference in the properties and total flavonoids of the water extract of Fruotus choerospondiatis,the content of gallic acid was significantly reduced(P<0.05),the number of bacterial colonies and the total number of fungal yeasts were significantly reduced(P<0.05).When the irradiation dose was 6kGy,the microbial level was in line with the provisions of the Chinese Pharmacopoeia 2020.Conclusion:Different doses of electron beam irradiation sterilization have no significant effect on the quality of Fruotus choerospondiatis,and good sterilization effect can be achieved when the irradiation dose reaches 6kGy.Electron accelerator irradiation is applicable to the sterilization of the seedless medicinal powder of Fruotus choerospondiatis,and has a significant impact on the quality of the water extract of Fruotus choerospondiatis,mainly reflected in reducing the content of gallic acid.
