雷公藤-甘草配伍干预肾病综合征大鼠AVPR2/AQP2的表达 被引量:1

Expression of AVPR2 and AQP2 in rats with nephrotic syndrome and the intervention of the compatibility of Tripterygium wilfordii and Glycyrrhiza uralensis

摘要 目的探讨阿霉素诱导的肾病综合征大鼠AVPR2和AQP2的表达以及雷公藤-甘草配伍的干预机制。方法选取成年健康雄性SD大鼠,随机选取10只作为空白组,其余大鼠均接受尾静脉注射给予6.5 mg·kg^(-1)的阿霉素诱导复制大鼠肾病综合征模型。并采用随机数字法将造模成功的大鼠分为4组(n=10):模型组(n=10)、阳性对照组(雷公藤多苷片80 mg·kg^(-1)·d^(-1),n=10)、雷公藤组(雷公藤提取物240 mg·kg^(-1)·d^(-1),n=10)及雷公藤-甘草组(雷公藤提取物240 mg·kg^(-1)·d^(-1)+甘草提取物80 mg·kg^(-1)·d^(-1),n=10)。测定大鼠24 h尿蛋白含量、血清白蛋白、尿素、肌酐水平、肾脏中AVPR2和AQP2 mRNA表达水平。结果大鼠经阿霉素造模后表现出大量蛋白尿、低蛋白血、高度水肿症候。相比于模型组,雷公藤及配伍给药组大鼠精神状态较好,体质量增加明显。与模型组相比,雷公藤组24 h尿蛋白、尿素、AQP2 mRNA表达均显著下降(P<0.05);雷公藤-甘草组24 h尿蛋白、血清白蛋白、尿素、肌酐、AVPR2 mRNA及蛋白、AQP2 mRNA及蛋白表达均显著回调(P<0.05)。相比于雷公藤组,雷公藤-甘草组的24 h尿蛋白、血清白蛋白、AVPR2 mRNA、AQP2蛋白表达回调更加显著(P<0.05)。结论雷公藤可显著降低肾病综合征大鼠AVPR2和AQP2 mRNA高表达,且与甘草配伍后效果更优,雷公藤-甘草配伍给药干预阿霉素诱导的肾病综合征大鼠的机制与纠正AVPR2与AQP2的高表达、减少水的重吸收有关。 Objective To explore the expressions of AVPR2 and AQP2 in rats with nephrotic syndrome and the intervention mechanism of the compatibility of Tripterygium wilfordii and Glycyrrhiza uralensis.Methods Ten adult male rats were randomly selected as blank groups,and the other rats received an intravenous injection of 6.5 mg·kg^(-1) adriamycin to induce nephropathy syndrome model.Rats with nephropathy syndrome were randomly divided into four groups(n=10):model group,positive control group(Regtatodoside tablets,80 mg·kg^(-1)·d^(-1)),Tripterygium wilfordii group(Regtovine extract,240 mg·kg^(-1)·d^(-1))and Tripterygium wilfordii and Glycyrrhiza uralensis group(Regtovine extract 240 mg·kg^(-1)·d^(-1)+liquorice extract 80 mg·kg^(-1)·d^(-1)).Blank group and model group were received saline(10 mL·kg^(-1)·d^(-1)).The contents of urine protein,serum albumin,urea,creatinine levels,AVPR2 and AQP2 mRNA levels were determined at 24 h.Results Rats with nephrotic syndrome showed high protein level in urine,low protein level in serum and high edema.Compared with the model group,rats of Tripterygium wilfordii group and Tripterygium wilfordii combined with Glycyrrhiza uralensis group showed better mental state and obvious increase of body weight.Compared with the model group,urine protein,urea and AQP2 mRNA levels significantly decreased at 24 h(P<0.05)in Tripterygium wilfordii group;The urine protein,serum albumin,urea,creatinine,expressions of AVPR2 mRNA and AVPR2 protein,AQP2 mRNA and AQP2 protein levels retuned obviously(P<0.05)at 24 h in Tripterygium wilfordii and Glycyrrhiza uralensis group.Compared to Tripterygium wilfordii group,24 h urine protein,expressions of AVPR2 mRNA and AQP2 protein decreased and serum albumin increased(P<0.05)in Tripterygium wilfordii and Glycyrrhiza uralensis group.Conclusion Tripterygium wilfordii can significantly reduce the high expressions of AVPR2 and AQP2 mRNA in rats with nephrotic syndrome,and obtain better effects by the compatibility with Glycyrrhiza uralensis.The mechanism of the compatibility of Tripterygium wilfordii and Glycyrrhiza uralensis antagonizing Adamycin-induced nephrotic syndrome is related to the decrease of the high expressions of AVPR2 and AQP2,and the water reabsorption.
