柴胡4种活性成分的体外抗氧化作用 被引量:2

摘要 目的评价柴胡4种活性成分体外抗氧化作用。方法通过紫外分光光度法,采用1,1-二苯基-2-三硝基苯肼(DPPH)和2,2-联氮-二(3-乙基-苯并噻唑-6-磺酸)二铵盐(ABTS)化学试剂法检测柴胡4种活性成分的清除自由基的作用;通过构建过氧化氢(H_(2)O_(2))诱导人正常肝细胞(LO2)损伤模型,给予柴胡多糖、黄酮、皂苷干预,测定模型细胞存活率、谷胱甘肽过氧化物酶(GSH-Px)、超氧化物歧化酶(SOD)活性及丙二醛(MDA)水平,比较柴胡3种活性成分抗氧化作用。结果柴胡多糖、黄酮、皂苷和挥发油对DPPH的半数抑制浓度(IC 50)分别为38.718μg·mL^(-1)、51.297μg·mL^(-1)、3.149 mg·mL^(-1)和2.426 g·mL^(-1);柴胡多糖、黄酮、皂苷和挥发油对ABTS的IC 50分别为130.931μg·mL^(-1)、351.484μg·mL^(-1)、5.422 mg·mL^(-1)和2.241 g·mL^(-1);柴胡皂苷(60μg·mL^(-1))可使氧化模型细胞存活率提高至67.47%(P<0.05),GSH-Px和SOD酶活性分别提高至17.06和52.05 U·mg^(-1)(P<0.05),MDA含量减少至2.11μmol·g^(-1)(P<0.05)。结论柴胡多糖、黄酮和皂苷具有体外抗氧化作用,可为柴胡临床用于疏肝解郁提供理论依据。 Objective To evaluate the antioxidant activity of four active ingredients from Bupleurum in vitro.Methods DPPH(1,1-diphenyl-2-trinitrophenyl hydrazine)and ABTS[2,2-biazo-bis(3-ethyl-benzothiazole-6-sulfonic acid)]were used to detect the free radical scavenging activity of four active ingredients of Bupleurum by UV spectrophotometry.The model of H_(2)O_(2)-induced injury in normal human hepatocytes(LO2)was established.The survival rate,GSH-Px,SOD activity,and MDA of the model cells were used as indexes to detect and screen the antioxidant effects of three active ingredients of Bupleurum.Results The clearance rates of DPPH by Bupleurum polysaccharides,Bupleurum flavonoids,Bupleurum saponins,and Bupleurum volatile oil were 38.718μg·mL^(-1),51.297μg·mL^(-1),3.149 mg·mL^(-1) and 2.426 g·mL^(-1),respectively.The clearance rates of ABTS by Bupleurum polysaccharides,Bupleurum flavonoids,Bupleurum saponins and Bupleurum volatile oil were 130.931μg·mL^(-1),351.484μg·mL^(-1),5.422 mg·mL^(-1) and 2.241 g·mL^(-1),respectively.Bupleurum saponins(60μg·mL^(-1))increased the cell viability to 67.47%(P<0.05),significantly increased the activities of GSH-Px and SOD to 17.06 U·mg^(-1) and 52.05 U·mg^(-1)(P<0.05),and significantly reduced MDA content to 2.11μmol·g^(-1)(P<0.05).Conclusion Bupleurum polysaccharides,flavonoids,and saponins have antioxidant effects in vitro,which provides a theoretical basis for the clinical application of Bupleurum in soothing the liver and relieving depression.
