摘要
目的:探讨长链非编码RNA CASC10在胃癌中的表达情况及对胃癌细胞增殖、迁移和侵袭能力的影响。方法:从GEO数据库选取胃癌数据集GSE50710和GSE65801并进行差异表达基因分析,使用GEPIA数据库预测胃癌中CASC10的表达情况及对患者预后的影响。利用qRT-PCr 技术检测CASC10在胃癌细胞和正常胃黏膜细胞中的表达差异,通过EdU、CCK-8和平板克隆实验检测胃癌细胞的增殖能力,利用细胞划痕和Transwell实验检测胃癌细胞的迁移和侵袭能力。结果:分析GEO和GEPIA数据库中数据集结果发现胃癌组织中CASC10高表达(P<0.01),并且CASC10高表达患者较低表达患者有更短的总生存(P<0.001)和无疾病生存时间(P<0.01)。CASC10在胃癌细胞系MKN-45、AGS、MKN-28、HGC-27中的表达较正常胃黏膜细胞GES-1升高(P<0.001)。在HGC-27细胞中,敲低CASC10后胃癌细胞的增殖、迁移和侵袭能力下降(P<0.05)。在MKN-45细胞中过表达CASC10后胃癌细胞的增殖、迁移和侵袭能力上升(P<0.05)。结论:CASC10在胃癌中高表达,并能促进胃癌细胞的增殖、迁移和侵袭,可作为胃癌潜在的分子治疗靶点。
Objective:To investigate the expression of long non-coding RNA CASC10 in gastric cancer (GC)and the effect on proliferation and invasion of GC cells.Methods:GC dataset GSE50710 and GSE65801 were selected from GEO database and analyzed for differential expression gene.Then the expression level of CASC10 in GC and the impact on prognosis were verified using dataset GEPIA.CASC10 expression in GC cell lines and normal gastric mucosa cell line were detected by qRT-PCr ,EdU,CCK-8 and CFA.Transwell and wound healing assays were used to determine the changes in proliferation,migration and invasion of GC cells.r esults:GEO and GEPIA dataset analysis revealed that CASC10 was upregulated in GC tissues(P<0.01),and the high expression of CASC10 was correlated with poor overall survival and disease-free survival in GC patients(P<0.001 or P<0.01).The expression level of GC cell MKN-45,AGS,MKN-28 and HGC-27 was higher than that of normal gastric mucosa cell GES-1(P<0.001).The cell proliferation,migration and invasion were inhibited in HGC27 cell after CASC10 knockdown(P<0.05),whereas CASC10 overexpression promoted the proliferation,migration and invasion in MKN-45 cells(P<0.05).Conclusion:CASC10 is over-expressed in GC,and may promote the proliferation,migration and invasion,suggesting that CASC10 can be a potential therapeutic target for GC.
