摘要
目的:探讨邻苯二甲酸二乙基己酯(DEHP)体内暴露对小鼠胚胎卵裂、囊胚形成、胚胎着床及胚胎线粒体膜电位的影响。方法:将40只3周龄清洁级ICR雌性小鼠(雌鼠)随机分为DEHP低剂量(300 mg/kg)组、DEHP中剂量(600 mg/kg)组、DEHP高剂量(1200 mg/kg)组和对照组,每组10只。采用灌胃方式构建DEHP暴露小鼠模型,按照5 mL/kg给药,每周5 d,每天1次,连续染毒8周。对照组灌胃玉米油。将各组雌鼠促排卵与非染毒雄性小鼠合笼后,取受精卵行胚胎培养,比较各组胚胎卵裂率、囊胚形成率、胚胎着床位点数和胚胎线粒体膜电位的差异。结果:DEHP低、中、高剂量组小鼠胚胎卵裂率、囊胚形成率及胚胎着床位点数均低于对照组(P<0.05),随着DEHP暴露浓度的升高,囊胚形成率下降(P<0.05);DEHP低、中、高剂量组小鼠胚胎线粒体膜电位均低于对照组(P<0.001)。结论:DEHP体内暴露对早期胚胎发育有毒性作用。
Aim:To investigate the effects of in vivo exposure to di-2-ethylhexyl phthalate(DEHP)on embryo cleavage,blastocyst formation,embryo implantation and mitochondrial membrane potential.Methods:Forty 3-week-old clean grade ICR female mice were allocated into DEHP low dose(300 mg/kg)group,DEHP medium dose(600 mg/kg)group,DEHP high dose(1200 mg/kg)group and the control group,with 10 mice in each group.The DEHP exposed mouse model was orally administered with DEHP once per day for 8 weeks(5 mL/kg,5 days per week).The control group was given corn oil.The ovulation induced female mice were caged with non-exposed male mice,and the zygotes were collected to compare the differences of embryo cleavage rate,blastocyst formation rate,implantation sites and embryonic mitochondrial membrane potential among the groups.Results:The embryo cleavage rate,blastocyst formation rate and embryo implantation sites of the DEHP low,medium and high dose groups were significantly lower than those of control group(P<0.05),and the blastocyst formation rate was significantly decreased with the increase of exposure dose(P<0.05);the mitochondrial membrane potential of embryo in the DEHP low,medium and high dose groups was significantly lower than that in the control group(P<0.001).Conclusion:Exposure to DEHP in vivo may has toxic effects on early embryonic development.
