摘要
原生质体是指利用一定方法去除细胞壁的裸露细胞,是一种无细胞壁的良好研究材料,在基因组学、转录组学、蛋白质组学、代谢组学和表观遗传学分析上均有广泛应用。原生质体培养体系的构建包括制备材料的选择和预处理、原生质体的分离和纯化、原生质体的培养与再生。牧草原生质体的相关研究较少,紫花苜蓿等常用牧草也尚未建立稳定的原生质体体系。本文对牧草原生质体分离纯化、培养再生及其应用的研究进展进行综述,旨在为牧草原生质体相关的研究提供参考。
Protoplasts refer to the bare cells with cell wall removed by certain methods.As a kind of good material without cell wall for research,protoplasts have been widely used in genomic,transcriptomic,proteomic,metabolomic and epigenetic analyses.The construction of protoplast culture system includes the selection of tissues for protoplasts and pretreatment of them,isolation and purification of protoplasts from the selected and pretreated tissues,and cultivation and regeneration of obtained protoplasts.Only a few studies on protoplasts of forage were conducted and even the main forage species,e.g.,alfalfa,are lack of stable and efficient protoplast culture systems.In this review,the progresses in isolation,purification,cultivation and regeneration of forage protoplasts and the applications of the obtained protoplasts were reviewed,aiming to provide a guide for the studies related to forage protoplasts.
