发光二极管630nm红光通过PI3K/AKT/mTOR通路抑制滑膜成纤维细胞炎症反应

摘要 发光二极管(Light Emitting Diode,LED)光照疗法是一种低能量光疗(Low-level Light Therapy,LLLT),在皮肤炎症治疗和术后疼痛缓解方面已被广泛研究,但其对类风湿性关节炎的作用及其机制尚不清楚。本研究采用LED 630 nm红光作为光源,以TNF-α刺激模拟炎症的类风湿关节炎患者组织中提取原代滑膜成纤维细胞(RA-FLS细胞)为模型。采用MTT和细胞划痕实验,检测不同剂量的LED 630 nm红光对炎症RA-FLS细胞的活力,及其增殖和迁移的影响。采用RT-qPCR和ELISA方法,检测IL-1β、IL-6和IL-8在细胞中mRNA表达及细胞上清中蛋白表达的变化。采用免疫印迹法,检测细胞中PI3K、AKT、mTOR和TRPV4蛋白表达和磷酸化的变化。结果证实,LED 630 nm红光可抑制RA-FLS细胞的增殖和迁移,降低细胞中IL-1β、IL-6和IL-8炎症因子的mRNA表达水平。光照剂量为39 J/cm^(2)的LED红光降低了RA-FLS细胞中PI3K、AKT、mTOR和TRPV4蛋白的表达和磷酸化水平。结果证实,LED 630 nm红光可以通过下调TRPV/PI3K/AKT/mTOR信号通路以及减少炎症因子的释放,抑制RA-FLS细胞的炎症反应及其增殖和迁移。 LED phototherapy,a form of low-level light therapy(LLLT),has been extensively studied for its therapeutic effect on skin inflammation and postoperative pain relief.However,its impact on rheumatoid arthritis and its underlying mechanisms remain unclear.In this study,LED 630 nm red light was used as the light source,and primary synovial fibroblasts(RA-FLS)were extracted from the tissues of rheumatoid arthritis(RA)patients,which were stimulated with TNF-ato simulate inflammation.The study employed MTT assay and cell scratch test to evaluate the viability of inflamed RA-FLS cells and the effects of different doses of LED 630 nm red light on their proliferation and migration.Changes in mRNA and cell supernatant levels of IL-1b,IL-6 and IL-8 were detected using RT-qPCR and ELISA.Changes in the expression of PI3K,AKT,mTOR and TRPV4 proteins in the cells were detected using western blot.The results showed that LED 630 nm red light could inhibit the proliferation and migratory effects of RA-FLS cells,reduce the mRNA and content expression of IL-1b,IL-6 and IL-8 inflammatory factors in cells.At the protein level,the effect of 39 J/cm^(2) of LED red light affected the expression and phosphorylation of PI3K,AKT,mTOR and TRPV4 proteins in RA-FLS cells.These results suggest that LED 630 nm red light can reduce inflammatory factor release and inhibit proliferation and migration in RA-FLS cells,and this effect may be related to the TRPV/PI3K/AKT/mTOR signaling pathway.Keywords:LED;630 nm red light;rheumatoid arthritis;PI3K/AKT/mTOR signaling pathway;synovial cells;low-Level light therapy.
