摘要
目的建立同时测定泽菊降脂片中绿原酸和橙黄决明素含量的方法。方法采用高效液相色谱法,色谱柱:Wondasil^(TM) C_(18)(4.6 mm×250 mm,5μm);流动相:0.4%磷酸水溶液-乙腈(梯度洗脱);体积流量:1.0 ml·min^(-1);检测波长:0~15.0 min,327 nm(绿原酸);15.0~40.0 min,286 nm(橙黄决明素);柱温:30℃,进样量:10μl。结果绿原酸和橙黄决明素检测质量浓度线性范围分别为7.61~76.08μg·ml^(-1)(r=0.9999)、3.12~31.16μg·ml^(-1)(r=0.9999);加样回收率分别为98.37%~101.37%(RSD=1.02%,n=9)、97.82%~101.20%(RSD=1.10%,n=9)。结论该方法简单准确、重复性好,适用于同时测定泽菊降脂片中绿原酸和橙黄决明素的含量。
Objective To establish an HPLC method for simultaneous determination of contents of chlorogenic acid and aurantio-obtusin in Zeju Jiangzhi tablets.Methods The HPLC method was adopted on a Wondasil^(TM) C_(18) column(4.6 mm×250 mm,5μm)with a mobile phase composed of acetonitrile-0.4%phosphoric acid in a gradient elution manner at the flow rate of 1.0 ml·min^(-1).The detection wavelength was set at 327 nm for chlorogenic acid in 0-15.0 min,and at 286 nm for aurantio-obtusin in 15.0-40.0 min.The column temperature was 30℃and injection volume was 10μl.Results The linear ranges of chlorogenic acid and aurantio-obtusin were 7.61-76.08μg·ml^(-1)(r=0.9999)and 3.12-31.16μg·ml^(-1)(r=0.9999),respectively.The recoveries were 98.37%-101.37%(RSD=1.02%,n=9)and 97.82%-101.20%(RSD=1.10%,n=9),respectively.Conclusion This method is simple,accurate and reproducible,and can be used for content determination of chlorogenic acid and aurantio-obtusin in Zeju Jiangzhi tablets.
