摘要
采用系统溶剂提取法制备了猕猴桃根石油醚、二氯甲烷、乙酸乙酯、正丁醇、水等5类提取物,并通过噻唑蓝法(methyl thiazolyl tetrazolium,MTT)评价它们对5种结肠癌细胞(SW480、HCT⁃116、HCT⁃15/Taxol、HCT⁃8/V、HCT⁃116/5⁃FU)和人正常结肠上皮细胞(FHC)的抑制作用。结果显示,猕猴桃根石油醚提取物对SW480、HCT⁃8/V细胞的作用最突出,IC50分别为(16.13±1.22)μg/mL、(21.61±1.46)μg/mL,且具有较高的选择指数(SISW480=16.93,SIHCT⁃8/V=12.64)。同时,采用液质联用技术从石油醚提取物中分析鉴定出了31种成分,其中包括20种萜类,5种香豆素和6种有机酸。
Five kinds of extracts from Actinidia chinensis Radix were prepared by systematic solvent extraction meth⁃od,including petroleum ether,dichloromethane,ethyl acetate,n⁃butanol and water.Subsquently,the inhibitory effects of the five extracts on SW480,HCT⁃116,HCT⁃15/Taxol,HCT⁃8/V,HCT⁃116/5⁃FU and FHC cells were evaluated by methyl thiazolyl tetrazolium(MTT).The results showed that the five extracts could inhibit the growth of 5 kinds of tumor cells(SW480,HCT⁃116,HCT⁃15/Taxol,HCT⁃8/V,HCT⁃116/5⁃FU).The petroleum ether extract had the most prominent effect on SW480 cells(IC50=16.13±1.22μg/mL)and HCT⁃8/V cells(IC50=21.61±1.46μg/mL)with an ideal selection index,respectively.Simultaneously,31 components,including 20 terpenes,5 coumarins and 6 or⁃ganic acids,were identified from the petroleum ether extract by HPLC⁃HRMS.
