摘要
结直肠癌(CRC)是目前全球范围内发病率第3的恶性肿瘤,具有治疗难度大、早期发现困难、易转移等特点。低糖、低氧、化疗等多种刺激因素能够使细胞出现内质网应激(ERS),应激细胞随后会产生未折叠蛋白反应(UPR)和自噬等纠正机制恢复细胞稳态,但持续过度的ERS则会导致细胞死亡。已有研究发现ERS参与CRC发生发展的不同阶段,促进或阻碍CRC的发生发展,两者关系密切。因此,从ERS介导的各条途径来探寻治疗CRC的新方案具有重要意义。文章将从ERS的作用机制及其在CRC中对肿瘤细胞增殖、凋亡、迁移、耐药、自噬等方面的作用进行综述,以期为CRC的防治研究提供新思路。
Colorectal cancer is the third most common malignant tumor in the world,which is difficult to treat,difficult to detect in the early stage,and easy to metastasize.Endoplasmic reticulum stress(ERS)can be induced by low glucose,low oxygen,and chemotherapy,and the stressed cells produce unfolded protein response(UPR)and autophagy to restore cell homeostasis,but excessive ERS will lead to cell death.It has been found that ERS is involved in different stages of CRC development,promoting or hindering the development of CRC,and there is close relationship between ERS and CRC.Therefore,it is of great significance to explore a new treatment scheme for CRC from the various pathways of ERS induction.This article reviews the mechanism of ERS and its effects on cell proliferation,apoptosis,migration,drug resistance,and autophagy in the treatment of CRC,so as to provide new ideas for the prevention and treatment of CRC.
