毛菊苣正丁醇萃取物中母菊素含量测定及其对大鼠肝纤维化保护作用

Determination of content of matricin extracted from Cichorium glandulosum Boiss et Huet with n-butanol and its protective effect on hepatic fibrosis in rats

摘要 目的研究毛菊苣正丁醇萃取部位(Cichorium glandulosum N-butanol extraction site,CGE)对大鼠肝纤维化(hepatic fibrosis,HF)的保护作用并测定其主要有效成分母菊素的含量。方法采用HPLC法测定CGE中母菊素的含量及方法学考察;大鼠随机分为正常组,模型组,CGE低、中、高剂量组,姜黄素组,除正常组大鼠背部sc生理盐水外,其余各组大鼠按体质量剂量sc 40%CCl 4橄榄油溶液建立HF模型,每周2次,连续12周,于第5周起,对相应给药组的大鼠在每日同一时间ig相应剂量的药物。12周后取肝脾组织及血清,检测肝脾指数及相关血清指标;观察肝脏病理组织学变化;采用免疫组化染色法和Western blot法考察肝组织TGF-β1、Smad3、Smad7及α-SMA的蛋白表达。结果母菊素在CGE中的含量为7.69%;模型组大鼠肝功能严重受损,出现炎症细胞浸润及纤维蛋白,提示HF模型建立成功;与模型组比较,各给药组可明显降低肝脾指数及血清中的AST、ALT、LDH、γ-GT、AKP和MDA水平,明显改善肝组织病理损伤程度,能够有效抑制HF大鼠肝组织的TGF-β1、Smad3、α-SMA的蛋白表达,上调Smad7蛋白表达。结论CGE主要成分是母菊素,CGE对CCl 4诱导的大鼠HF具有保护作用,其作用机制可能与阻断TGF-β1/Smads信号转导通路有关。 Aim To investigate the effects of Cichorium glandulosum N-butanol extraction site(CGE)on hepatic fibrosis(HF)in SD rats and to determine the content of the main effective component matricin.Methods HPLC method was used to determine the content of matricin in CGE.The SD rats were randomly divided into control group,model group,CGE low-dose groups,medium-dose and high-dose,and curcumin group.In addition to control group rats′back subcutaneous injection(sc)normal saline,rats in the other groups were treated with body weight sc 40%CCl 4 olive oil solution to establish HF model,twice a week for 12 consecutive weeks.From the fifth week,rats in the corresponding administration groups were given the corresponding doses intragastrically at the same time every day.After 12 weeks,liver and spleen tissues and serum were taken to detect liver and spleen index and related serum indexes;the histopathological changes of liver were observed.The protein expressions of TGF-β1,Smad3,Smad7 andα-SMA in liver tissue were detected by immunohistochemical staining and Western blot.Results The content of matricin in CGE was 7.69%.In the model group,the liver function of rats was severely damaged,and inflammatory cell infiltration and fibrin appeared,indicating that HF model was successfully established.Compared with the model group,all drug administration groups significantly reduced the liver and spleen index and droped out the levels of AST,ALT,LDH,γ-GT,AKP and MDA in serum,significantly improved the pathological damage degree of liver tissue,effectively inhibited the protein expression of TGF-β1,Smad3 andα-SMA in liver tissue of HF rats,and up-regulated the expression of Smad7 protein.Conclusions The main component of CGE is matricin,which has a significant protective effect for CGE on CCl 4-induced liver fibrosis in rats,and its mechanism may be related to blocking TGF-β1/Smads signal transduction pathway.
