摘要
目的探讨何首乌在九蒸九晒和九蒸九烘干燥工艺中,其外观性状、浸出物和化学成分的变化规律。方法分别采用九蒸九晒和九蒸九烘炮制工艺制备样品,观察外观性状;参考2020年版《中国药典(四部)》通则2201浸出物测定法测定醇溶性浸出物;建立不同炮制样品的高效液相色谱指纹图谱,以二苯乙烯苷峰为参照峰,导入中药色谱指纹图谱相似度评价系统(2012.130723版),鉴定其中的化学成分。结果何首乌炮制后饮片颜色逐渐加深,显黑褐色或棕褐色,质硬,断面角质样,第9次炮制后有明显蜡样光泽,晒干品和烘干品无明显差异;醇溶性浸出物在2种炮制过程中均逐渐减少并趋于平稳。生品和炮制品的高效液相色谱图中共检出22个峰,其中7个共有峰,指认出没食子酸、5-羟甲基糠醛、二苯乙烯苷、大黄素、大黄素甲醚,发现7个新成分。3批炮制品中,第1批烘品中5种成分的含量比晒品稍高,第2批和第3批晒品中5种成分的含量比烘品稍高。结论何首乌经上述2种工艺炮制后的结果无显著差异,可考虑用九蒸九烘的现代工艺代替传统的九蒸九晒工艺。
Objective To investigate the changes in appearance,extract,and chemical components of Polygoni Multiflori Radix processed by nine-time repeat of the steaming and sun-drying process and nine-time repeat of the steaming and oven-drying process.Methods Samples were processed by nine-time repeat of the steaming and sun-drying process and nine-time repeat of the steaming and oven-drying process techniques,and their appearance characteristics were observed.According to general rule 2201 of the Chinese Pharmacopoeia(2020 Edition,VolumeⅣ),the determination of alcohol-soluble extracts was carried out.The high-performance liquid chromatography(HPLC)fingerprints of different processed samples was established with the stilbene glycoside peak as the reference peak,and the Similarity Evaluation System for Chromatographic Fingerprint of Traditional Chinese Medicine(Version 2012.130723)was used to identify the chemical components.Results The color of the processed slices of Polygoni Multiflori Radix gradually deepened,appearing black brown or sepia,with a hard texture and a horny cross-section.After the 9th processing,it had a significant wax-like luster,and there was no significant difference between the sun-dried and oven-dried products.The alcohol-soluble extract of Polygoni Multiflori Radix showed a trend of decreasing first and then stabilizing.A total of twenty-two peaks were detected in the HPLC chromatograms of the raw and processed products,including seven common peaks,the gallic acid,5-hydroxymethylfurfural,diphenyl ethylene glycoside,emodin,and physcion were identified,and seven new components were found.Among the three batches of processed products,the content of five components in the first batch of oven-dried products was slightly higher than that in the sun-dried products,while the content of five components in the second and third batches of sun-dried products was slightly higher than that in the oven-dried products.Conclusion There is no significant difference in the results of Polygoni Multiflori Radix processed by the above two processes.Therefore,the modern process of nine-time repeat of the steaming and oven-drying process can be considered to replace the traditional nine-time repeat of the steaming and sun-drying process.
