摘要
高压蒸气灭菌是高等级生物安全实验室废弃物处理的重要手段。为了探索不同种类废弃物[防护服、生物安全柜废弃物(手套、吸水垫)、小鼠笼盒、容器类废弃物(离心管、细胞瓶、利器盒)]的有效灭菌模式,试验将防护服(设置上、中、下三个点位)和生物安全柜废弃物分别进行1,2,3层包装,小鼠笼盒和容器类废弃物分别进行1,2层包装,分别采用固体灭菌程序[分为排空(15~80 kPa,循环3次)、升温、灭菌(121℃,30 min)、排气等阶段]和织物灭菌程序[分为预热、脉动(真空-80~80 kPa,循环3次)、升温、灭菌(121℃,30 min)、排气、干燥等阶段]进行高压蒸气灭菌,并分别通过生物指示剂及化学指示卡对灭菌效果进行评价。结果表明:固体灭菌程序可对1层包装的防护服上点位进行有效灭菌;对1层包装的生物安全柜废弃物(手套、吸水垫)、小鼠笼盒、容器类废弃物(离心管、细胞瓶、利器盒)的灭菌效果优于多层包装,但容器类废弃物由于本身容器的强密封性,即使是1层包装蒸气也很难进入,灭菌效果并不理想。织物灭菌程序对不同包装层数的所有种类废弃物均可有效灭菌。化学指示卡与生物指示剂检测结果差异不明显。说明对于高等级生物安全实验室废弃物,织物灭菌程序(脉动真空灭菌模式)的灭菌效果优于固体灭菌程序(单纯机械排空灭菌模式),且更加适合多层包装的多种废弃物同载灭菌。
Autoclave sterilization is an important means of waste disposal in high level biosafety laboratory.In order to explore the appropriate sterilization mode for different kinds of waste(protective clothing,biosafety cabinet waste[gloves,absorbent pads],mouse cage box and container waste[centrifugal tube,cell bottle,sharp tool box]),in this experiment,protective clothing(upper,middle and lower three points)and biological safety cabinet waste were packaged in 1,2 and 3 layers,respectively,and mouse cage boxes and container waste were packaged in 1 and 2 layers,respectively,which were autoclaved by the solid sterilization procedure(containing emptying[15 to 80 kPa,3 cycles],heating,sterilization(121℃,30 min),exhaust and other stages ] and fabric sterilization procedure(containing preheating,pulsation[vacuum-80 to 80 kPa,cycle 3 times],heating,sterilization[121℃,30 min],exhaust,drying and other stages),respectively.And the sterilization effect was evaluated by biological indicator and chemical indicator card,respectively.The results showed that the solid sterilization procedure could effectively sterilize the spots on the protective clothing with one-layer packaging.The sterilization effect of one-layer biosafety cabinet wastes(gloves,absorbant pads),mouse cage boxes,and container wastes(centrifugal tubes,cell bottles,and sharp tool boxes)was better than that of multi-layer packaging.However,due to the strong tightness of the container itself,it was difficult for vapors to enter even one-layer packaging,and the sterilization effect was not ideal.Fabric sterilization procedures could effectively sterilize all types of waste with different packing layers.There was no significant difference between the results of chemical indicator card and biological indicator.The results indicated that the sterilization effect of fabric sterilization procedure(pulsating vacuum sterilization mode)was better than that of solid sterilization procedure(simple mechanical empting sterilization mode)for high-level biosafety laboratory wastes,and it was more suitable for the sterilization of multiple wastes in multi-layer packaging.
